Bad inoculum? or bad inoculation technique?

Hi everyone,

My friend has recently inoculated several grain & PF jars but has not had great success rates.

He's used the same inoculum for all jars (colonized dextrose water) and some worked and some didn't (green mould mostly). It's not the substrate jars themselves as they stayed uncolonized by anything for over a week until inoculation (that and he PC's for 1.5 hours generally)

This indicates that his inoculation technique needs work right? if it was the inoculum then all jars would be contaminated.. correct?

Can anyone confirm this?

step thru your process. Where did you lack a little effort. Were syringes sterile before adding innoculant? did you alky rub down everything before injecting?

maybe PC for a little longer and try again.

I don't think you can rule out the innoculum, either...

Liquid cultures can be difficult, and hard to diagnose as contaminated. Any non-white non-stringy growth in a LC jar is trouble. Be on the lookout for floating particles when you shake the jar.
