Mycelium Growth on Red Agar, ID please?
    #25157617 - 04/22/18 12:26 PM (6 years, 8 months ago)

Hello World! This is my first post here but I've been reading for a while.

I obtained a spore print through friends, said to be PC Burma.

I decided to go from print to agar.

Red Agar-Agar and Potato Flake boiled down into Mini Rounds.
I was supposed to add honey and even had it right next to the pot but left out a drop!
I read into others' inquiries about leaving it out and while it seems highly desirable to promote growth, the lack of glucose only lends to a longer wait period ( as well as more time for bacteria growth to overtake the spores/mycelium).

I used sterilized swabs from print to agar in a Z shape. About three different plates.

So I waited about a week and a half before seeing growth.

At the first sign of growth I transferred to an unopened/sterilized agar "plate".

Days later I transferred another primary plate's first growth to a secondary plate as well.

Now, this is my first time and I'm unsure if I'm simply looking at entirely contaminated plates or reasonable growth.

My apologies for the picture quality but I'm ready to take more if necessary. Bear in mind that these were taken at the same time, and not at the time of transfer.

A:

A1:

B:

B1:

C: I didn't cut from, just let growth continue:

D: May have split the slice from B?

TL;DR
I'm a noob. Made Agar, left out honey so I was waiting a while. Did a couple transfers.
Start over?

ALSO, it seems to be discoloring the Agar! what's up with that?

The last picture seems good to me but definitely get more opinions.
A1 could also be OK
B1 I have the suspicion that it could be trich mold in a few spots but maybe its the picture/flash/reflection I dunno.

the mycelium is eating the agar with food coloring in it so as it digests the agar the coloring gets digested also

They look like there's bacteria in them. I'd try making some more new plates. I wouldn't trust these ones.

I think A1 and B1 look ok to me, but were the A, B and C made with the same agar? They have a “waxy bacteria” look, in my opinion.

Just because I'm reluctant to trash everything and start over I've taken more pictures:

The darker yellowish/greenish parts look sketch.  Take the white mycelium and transfer to new plates.

Original A1 looks good. B1, not so sure.

If it were me I'd grab transfers from that first pic in the new set of pics from the 11 to 1 o'clock position. And maybe the third one from 1 to 3 o'clock area. Definitely agree to stay away from the disorganized and discoloured shit.

Yeah the overgrown plates shouldn't get any xfers. They may pin actually and you could grab a clone  I've done it before.

Hell yeah! Thanks everyone.

The discoloration is simply from a shallow pour combined with the flash through the transparent mini rounds. I actually only see white, I've only been concerned over the texture. One of the overgrown plates I just tossed actually had some rhizomorphic growth over the fuzz. Unfortunately I noticed as it went into the trash. Either way, I need more plates!

I'll prep more agar with some actual glucose and report back after I've done another transfer.

I plan to grain spawn in solely WBS as I haven't found any whole oats locally. Then bulk spawning on Coco Coir, perhaps with a tiny bit of spent coffee grounds [boiled through to remove excess nitrogen] ? Personal opinions on gypsum?

Thanks again!
