Hey, made some pasty plates and I used very bad sterile technique lol, but is this plate contaminated completely, or am I able to transfer the clean parts (if there are any) to another plate?

That was inoculated with a syringe, cooked em at 10-11 psi for an hour, used a SAB and got all contams \: I used the wrong kind of tape tho ahaha, it was like silicon removal tape lmao. Those two actually didn't go thru the PC and looked the best lol

Those contamination rates make me wonder if something is off with your process.

I use Petri dishes and pour the agar into the petri dish in an sab myself

Yeah I used the easy agar tek, but used square ones instead, but that seems like its not a problem. I PC'd them for 1 hour in an electric one that only goes to like 10-11 psi, also I opened them up several times lol.

Either you didn't sterilize the plate I guess ? Or introduced a contamination.
I'm not sure what pressure they need to reach for how long. I thought I did mine for like 60 min at 15psi but idr, been a while since I've done plates and I won't need to for a while. I would have to look it up.
Possibly checking on it didn't allow the right pressure for the correct duration, I'm sure that could be a problem.
I would recommend using a SAB if you don't use one

Nah like opening them as they colonized, I used a SAB and cleaned out a bathroom, idk if you are supposed to vent with am electric one, gettin the stove top one soon so I'll probably have much better success lol

His Tek says40-45 min I didn't see what psi.
Opening while colonizing? Yeah bro that's a definite no go
Vent with electric one?
You'll probably have much better luck if you don't open it until youre in your sab ready to use it

yeah theres like a sealing and venting latch, i put it on the venting setting while it got up to pressure, then cooked for an hour at like 10 psi, his tek was 15 psi for 40-45 min, so i compensated pressure for time. Don't open the agar plates till your ready to inoculate with them you mean right? and yeah I'm just gonna tape em shut next time lol, also gonna have a 23 qt presto stove top, so I'm sure it'll work much better lol
