Dont know what im looking at on Agar
    #25144541 - 04/16/18 10:36 PM (6 years, 8 months ago)

Im new to Agar. I havent got any grows started from any plates yet.  The first time I did agar I used Pastywhyte's no pour tek. I have no Idea if this is cob web mold or if this is healthy mycelium.

I didnt like the fact that I couldnt see very well with the no pour tek so I stepped up my game after this and now I pour my own into petri dish's.
I still dont know what Im looking at though. Is this healthy mycelium is there anything worth transferring here?

These are all from a spore syringe's.

I'm sorry to say, that is a lot of bacteria.

Edit: at least on those plates.

Yes, thats bacteria on petri dish, the no pour one was lookin better. If you wanna start from agar, i highly suggest to go with a print, i got the same issue with syringe, on agar we dont need the spore to be in liquid, liquid just add another thing that can go wrong imo.

Good luck.

thank you for the info. I will throw out the petri dishes. So the no pour tek is mycelium? its so hard to tell out of those unclear things.

Did you pour those plates in a SAB? Did you start from syringe? I’m a noob too, just got started with agar a few weeks ago. Im sure my sterile tek is mediocre at best as i just got started, but as of now ive only had 2 plates out of 20 contam with mold. Im in the process of trying to save contaminated jars (bacteria) with agar work and having relative success using standard SAB procedures. Your petris definitely look like bacteria. Good Luck!

Like you said it's unclear. But did you inoculate them with the same syringe ? Cause your petri look pretty much like mine was, i tried 5 syringes from the same source and i never got even a tiny mycelium grow out of these bacterial festival syringes. But well, from what i can see in the no pour, it may have some mycelium in there, but its hard to confirm with that vision for me, and im still not really experimented, but with bacterial syringe, i have some experience lol

Yes the no pour tek and those two petri dishes are from the same spore syringe. I dont think im going to chance using any of those. I just did 3 new petri dishes with a spore print I took a couple weeks ago.

SebQc Those pics of petri dishes are the ones that are bacterial? because those look exactly like the ones I posted pics of.

That stuff in those dishes is straight bacteria characterized by it's greasy appearance.

You could probably snag a bit of mycelium out of that no pour dish and transfer to a new one if you don't want to wait for germination again, just be sure to make a few dishes in case you lose some. You can take a tiny transfer if you see a clean spot but still you're going to have to clean that culture up a bit. Don't worry about taking too small of a transfer either, iv'e transferred pieces of mycelium so small that I couldn't tell if there was even any on the receiving plate, they'll take a while to grow out like 7 - 14 days but it'll work. Let the new dishes mycelium grow away from the bacteria and grab a piece from the leading edge once it's surpassed the contaminant. Also when starting from spores i like to grab my first transfer when the mycelium patch is about the size of a dime, the sooner the better imo just because syringes are so unreliable for cleanliness. When I first started I ran 33 dishes off 11 syringes and had a bacteria party in my plates, it was bumpin' everyone was there! My spore syringe source is of a somewhat local origin and I don't think they have the best sterile technique lol.
