Grow log - Bods tub tek

Ok, so a little back story, I'll keep it brief.
Ive tried 6 un modded tub grows and have had sub standard results with each one. The common issue is yellowing (bacteria) and stalling. and side pins. which I assume are related. so I'm starting this grow log in order to share with you more experienced folk and maybe I can see where I'm going wrong along the way.
All of my previous tubs have been scrapped and I'm starting with these, that i have laying around

The 3 brown plates are grain water agar with spores looped on from a print I took a while back. These are fresh plates, maybe a week old?, and it looks like i can start taking some of those leading edges and transferring to begin the isolation process. please do correct me if im wrong.
the two lighter plates are BRF agar and these were partial isolates i had stored in the fridge from Nov. Not sure if these are usable. welcome to advice here
also have two grain jars colonized that i may transfer g2g and start a tub to have while my agar work is progressing.
Any thoughts on any of this so far, is welcome. And thanks in advance for any help and corrections.

Ok, so a quick update. Took the two existing jars and made a tub out of them. (normally id use 3 on a tub this size but they're a bit more full than ⅔ and they're all i have anyway) Noticed there was some liquid in the bottom of the jars as i was rolling them around. Not sure if thats due to being stored in the fridge or what.
cooked 325g coir (burpee) for 7-8 hrs w 2 ¼ quarts of water. mixed in tub, covered and put under the light. now i wait. will post pics as things progress. feel free to comment or correct me if you see anything suspect. thanks all.

Yeah, every tub I've tried has been slow compared to the timelines I've seen online and almost always ends up bacterial. It's the reason I'm showing everything step by step here to isolate where I'm going wrong. The only thing i can think of is my grain boiling time/technique is off or its something in the genetics? (if thats even possible?) We'll see i guess, once my agar work is ready to head to grain. Hard to say where i went wrong with these if they end up bacterial since they were done with an older batch of agar (my first try)

True. For me anyway. I mix my most recent tub on the 21st and by the 26th it was 100% colonized.

So, yeah...day 14 and its shown virtually no progress. Im assuming its fighting bacteria as that has been the routine with my tubs thus far. Guess I'll let it sit until i have something else to wait on, maybe it'll come through.

on to more promising results...

getting ready to take more samples from these plates. these are the first samples I've cut, transferred, and grown out, getting ready to do it again here. I'm thinking that im liking 3 the most. Looks a little thicker and and has been moving quickly. 5 is a close second. going to take these and make transfers today. Any suggestions, moving forward? im going to take smaller samples for the transfers next time. Any advice oh how many i should do? multiples from each plate? or one from each? what do you experienced guys/gals do here? thanks!

You can do plates any way you want so long as clean mycelium ends up in your spawn.
Something is up with that tub, at 14 days it looks like it should at day two still. I would guess bacteria as well.
All 5 plates have stuff i would use. They all look pretty good

From what I see I like number 5 at around 9 O'clock and number 4 at 6 O'clock too. Can't wait to start some agar! Best of luck and pray you get a good mono going soon!

plates are growing out well, it seems. I'm thinking i need more transfers before going to grain? what areas look most desirable? I took a guess the last time i went to grain and im not sure i did it correctly. open to and advice or critique as usual. thanks!

I'd transfer once or twice still from the rhizo areas. like that third plate 4 o'clock.
Maybe flip the lid on the tub you have going, try to get more FAE

i just pulled out the grains for jarring and noticed a bunch of bugs on the towel beneath my colander. this is something ive been overlooking since the beginning. could this be the reason for stalled bacterial tubs. Should i even bother putting agar to these jars or just chuck the bag of grain, go get a new one and start over?

Shouldn't be an issue the grain gets boiled then sterilized anyway
Bugs are indicative of cleanliness and sanitation problems, not to say I do not get bugs in my house but i damn well try to stay on top off the bug situation.

You mentioned that the tub may be the problem. Are there any cracks on the bottom of sides of the tub? The foam moisture seal under the lid is it completely dry?

? I'm not sure i mentioned anything about the tub itself being an issue. If i did, it was purely by accident. none of my tubs have any holes or cracks and lids are all in good shape and dry aside from the condensation inside while colonizing. the tub in the pictures above never progressed and ended up sprouting a nice gnarly green spot. So, he's a goner and has been disposed of.

Ok, its been awhile. Ive been keeping tabs, just not posting. life happens. Anyhow, i took an agar plate to 10 grain jars and it took them about two weeks to fully colonize and tonight i used 5 of them for a tub.

Day 5, ( now, were talking!) never had a tub colonize this quickly. Not counting my chickens just yet, but it appears I may have solved the problem.
Things I did differently this time:
-New/fresh agar sample -left grain jars in pressure cooker overnight to cool down - used less water when cooking the coir in bucket tek (4.25 qt)
And i think posting this may have made me a bit more diligent with instructions, sterility and technique in the SAB. lets hope it all works out. I'll continue to post more progress pics as things move along.

Isolating from spores is kinda pointless unless you're going to test all the different isolates.
Better to do a small scale grow (even an in-vitro pp5 container or something, or a shoebox or whatever), and pick a shroom to clone. Then you at least know you have good fruiting genes.
Otherwise you could easily end up with an isolate that's a shitty fruiter.

Im just trying to stick with one tek until I can get good consistent results. Ive got time invested in this tek and I'm determined to sort it out. I haven't even read into cloning yet. But I'll get there.
