Need help using a vertical laminar flow hood.

I purchased this Vertical Laminar Flow hood and am wondering if i am at a loss now. Will i still be able to perform g2g transfers and culture work inside this unit properly?

i am learning now All the pros say stay way from the vertical flow, however i believe with the proper sterile technique, work should be a breeze.

I knew at least 1 person that ran a smaller commercial edible mushroom farm and uses a vertical hood with no problems. It's not something I would purposefully do but it can be used.

Do i just have to be careful to stay downwind of the flow and not work above exposed media? and make sure glove and arms are covered and sterile?

The air flow is laminar mostly despite some turbulence created from grouping jars to one side.  i was thinking about elevating my work so air may pass underneath. like if i keep cleaner stuff to the back of the hood and don't introduce contams behind or above the clean/open jars i should be ok? the flow will push the contams down onto the work surface and out

I believe its fine.  I've worked inside of units like that, albeit in a different, but not less sensitive, setting.

I believe your largest issue lies in ensuring your hands and arms are as clean as possible; sterile gloves and sleeves would be excellent in this case.  With horizontal flow, contaminants on your hands and arms tend to get blown back outward toward you.  With vertical flow they will be blown downward into your work area.  It looks like that hood has an air re-circulation mechanism that helps to pull "dirty" air out of the workspace, which is good, but don't ever block the grate openings on the bottom tray as this will render that mechanism useless.

I don't think you will have to elevate anything. You are just going to have to be more mindful of where your hands are and keep them from being directly over any open media. Like hold a jar towards the bottom when GTG instead of right at the neck.

It can work but as was said, you're gonna have to be super careful. If it were me, I'd sell it off and build/buy a horizontal hood.

So as long as I work off to the side or in front of the exposed media, contams should be forced down and out toward me mostly; because of the L shape air flow.

Just do not allow jars / hands do directly pass over the downward airflow directly over the open jar/Petri?

If I was you I'd look for some instructional videos on youtube. There are several. It's going to be something that you will have to practice and learn from your mistakes. Even if it was horizontal.

Honestly man I’ve only ever used horizontal flow. But yes definitely never have anything over exposed media/grains. Whatever is left on you from will blow directly down on your project and that’s bad juju. Please keep us updated with whatever projects you have though. I’d love to see you succeed and post it to share the knowledge. And even if something fucks up keep us posted so we can learn from each other’s mistakes. This entire forum is nearly two decades of trial and error.

The benches air flow has me thinking that as long as i keep my hands and arms inside the working area on the table then the contaminants will never have a chance to get to the left and right  areas of the table where my grain jars are pending.  And as long as i do not raise my hands or arms above the jars and keep from behind the working area i should be good.

i will be using tyvek sleeves and also gloves for the process.

spawn bags might be more tricky with this setup

this is how i look at the work area on the table.. like a 3 point line .. stay in that area and tams will be swept away from everything.

In a few days ill do some transfers... maybe even make up some plates tonight just to test the setup out...

ill definitely keep this updated with the VFH's success

Im thinking i can get it to  work, although it may not be ideal.

Next investment a horizontal bench?

If you decide to go horizontal I highly highly recommend the Baker EdgeGARD horizontal flowhoods. You wouldn't want to step down from the beautiful commercial hood you have to some ghetto DIY flowhood.

You oughta be doing several agar pours to test this thing before you do any critical work. Just pour some sleeves and see if you get any contams, then you'll know for sure.

Thinking my next purchase will be multiple flow hoods like the ones off of FP. line them up in a series along the wall with a work surface and have it an open setup without and enclosure

Proud to say that the agar plates i poured in the hood came back all clean.  I also performed a few open air inoculations and grain transfers in the hood. 

Vertical Flow Hoods can be used!!!.. despite what others like to believe..  it may require a bit more technique but overall i love my hood and it is performing as desired.

Nobody said they can't be used. I mean really you could probably pour agar in open air and get away with it. It's just not that reliable. Same with vertical flow hoods.

This looks like a great find. There are only minor differences in horizontal vs. vertical flow hoods, except Stamets happens to favor horizontal ones so now nobody will use anything else.

The technique is the same: wipe things with isopropanol before you put them in the hood, put clean things upstream from relatively dirty ones, and don't hover over open media.

The specs on your model are good and it has some nice features like a polypropylene base (you have a steel base), a water column meter to measure the HEPA lifetime, overhead lights, grounded outlets, etc. Do you also have a UV light?

Anyway, great purchase if you got it used, still better than Fungi Perfecti if new. Lots of vertical hoods use smaller filters, so you could cheaply upgrade to an ULPA filter when you replace the HEPA in a few years.

I wouldn't try using a vertical one for tissue culture work at all. Maybe simple beverage QA work but for tissue culture work its going to be a pain in the ass.
