Last night at work I had a hankering for spore knowledge of the agaricales and found a pretty good review article. It didn't give me the info I wanted but I thought someone here may find it interesting.

Apparently spores from Psilocybe mutans can survive 9 years (p.95 paragraph 4). There are many factors that contribute to spore germination, light/lack of light, heat or cold shock, amyloidity, and others that likely don't apply to cube spores.
The amyloid coat in particular has me interested. I've heard people on here mention adding water to the agar plate after inoculating spore solution. Does anyone know if Pislocybe spores carry an amyloid coat?
Other than that I've been trying to find research I could possibly apply to cubes, which the closest relative I can find with extensive research is Coprinopsis cinerea. It's a model organism for basidiomycetes' meiosis and mating type studies. If I find anything genetically interesting I'll try to compare it to whatever cubensis genome they have on NCBI.
Anyway I'm drunk and need to go to bed.

With the insane genetic diversity of fungi you arent going to be able to apply knowledge of one organism to another. Also, a lot of what you just posted is,over the head of many of us here. And I've been sober almost a week.

I´d like all the knowledgeable growers to chime in I need to know before I start doing some experiments. The poll to be as correct as possible I´d like only those that feel fairly comfortable answering to take the poll. If you´re not 100% sure but are pretty sure that it is in a certain range then go ahead and chime in please. Thanks guys

Being eukaryotes, they shouldn't differ by as much as you would think. And in the meiosis and mating type studies researchers have found that genes encoding for these processes are highly conserved across species.
I've learned so much from many of you here while I didn't post. I guess I just want to be able to give back in some way. Since I still suck too much at cultivation to innovate, figured I'd stick with what I know.

Sometimes when I see narrowed down MS genetics the crops remind me more of a clone. I guess the more strains involved the more variety in the tub and the fewer strains involved equal more uniform fruiting. And between a MS grow with huge variety and an isolated mono culture is just a spectrum I guess..

I made some Pasty plates but dripped directly from syringe (spore and DIY liquid culture) to agar instead of using a loop or swab. Guess I should have read a bit more, haha, but if this works, I'm not gonna complain. Next time, I'll probably use tools, but I got my nervousness about working with agar and SABs out of the way- I'm not sure why, but it's kinda exciting working in an SAB.

No worries sangreen, if there are viable spores they should still germinate! Just make sure to transfer healthy mycellium from it next time.

Yep, I'm expecting either growth full of contams (lol, worst case scenario) or wonky off-center growth. Nothing ventured, nothing gained- every mistake is a learning experience.

were you using agar? There's no need to filter water at all, let alone post full colonisation
