AGAR QUESTION

Hey guys,
I just watched this video: and would like to try agar for the first time. I'm planning on using agar agar powder, barley malt extract and nutrition yeast. However, I'm paranoid that I'm going to order something that will not work for agar. Will these three items I have linked work for making agar? The nutition yeast has a powder and flake version and I'm not sure which one to get. ALso not sure if the barley malt powder linked will work either. Thanks for any and all advice!

Looks good. You can make agar media with a variety of nutrients sources...dog food, potatoes, manure, etc. If you have a home brew store in your area get the malt extract there and save $10.

I dunno if I just got a really shitty brand. But I once bought a "malted barley extract" and for some reason the agar wouldn't set and the powder wouldn't mix in with it. It was the most bizar thing I'd ever seen. The powder kinda seperated from the water. Like when you see silt in a pond or something, I'm not describing it very well but the barley powder wouldn't mix well.
As mycolorado said there are a lot of things that can be used for nutrition.

Already have a SAB and PC along with some MS syringes so I should be able to have my first agar attempt in the next week. I will be using Bod's Comprehensive Agar Tek.

Will I be able to inoculate the agar with the ms directly after pouring all the agar in the petri dishes? If I have petri dishes ready in the fridge when I take them out can I immediately inoculate those as well or does it need to be turned back into a liquid?

You can use a ms to knock some agar up. For the other questions I will leave to someone else.

Don't put petris in the fridge unless you're storing cultures. They stay good just fine at room temp.
That being said yeah you can inoculate them whenever. After taking them out it should only take them an hour or two to warm up.

If I'm understanding you correctly, you want to wait until your agar has solidified and cooled before inoculating.
Right after it comes out of the fridge is fine.
To clarify... I wouldn't ever inoculate agar with spores when it's still warm enough to be a liquid.

You want dry malt extract (most of which is barley) not liquid malt extract, which will throw off your measurements, and also usually contains hops extract, which might have adverse affects.
Tho, I think hops is used as a preservative, so there's a chance it could have an anti-bacterial effect, but I would leave that experiment to people with more experience. It could also have an anti-fungal effect, which you probably don't want.

ok so i have my agar made and in the PC at the moment. im going to put the whole bottle in the fridge when i take it out. just to make sure I understand correctly, say i take it out of the fridge tomorrow. to warm it up i can microwave it a few mins at a time while shaking it in between until its liquid. after its liquid, get my SAB and inside I can pour agar in the petri dishes. at this point, I leave the petri dishes outside the fridge until they are not a liquid anymore and then i can go ahead and inoculate them after they are not warm anymore?

Watch it in the microwave because agar boils up really quick and easily, but yeah that's the process.

I put the petri dishes in the fridge immediately after inoculating. This shouldn't be an issue should it? Or should I store them outside the fridge until they show signs of germination?

They will not germinate or grow in the fridge. Why are you wanting yo put your petris in the fridge?

You only store un-poured agar in the fridge. Not plates, inoculated or otherwise.
You won't see any growth in the fridge.

I store all my MEA at room temp. Unpoured pyrex bottle full, 30 dishes poured and individually wrapped and ready to use. After pouring dishes let the agar solidify before inoculation. Once a dish is fully germinated than it can be stored in fridge for later use. I always wrap my dishes with glad cling wrap, used or unused. Try to always keep them sealed unless using in your SAB. Practice pouring dishes, that will get you used to the different consistancy and temps for agar use. It will also help your sterile technique. Contaminates will grow if you get any, even without MS inoculation. Help you to handle dishes and all. The agar will only be liquid while boiling and mixing it up for PC, and for pouring. To microwave it i do 30secs at a time in a 1100watt microwave. Shake in between every 30 seconds. It gets hot, keep lid on tight till ready to pour. Pay attention to bods details in his tek. I used his agar tek and its very simple, he gives very good advise and tips. Practice and play, it becomes very fun, and its the easiest!! Have fun and best luck!!

I thought I read somewhere that they would be fine in the fridge and that it would just take longer to germinate but maybe I was mistaken. I took them out of the fridge this morning, so the inoculated dishes were in there for a total of 12 hours. This shouldn't be an issue, should it?

Should be fine. Im pretty sure the fridge temperature doesnt kill, just slows down a lot. It may take up to 2 weeks for growth to show coming from the MS syringe.

Unused agar can be stored in the fridge, bottle or dishes. I have never had an issue with seperation or caramilzation or anything at room temperature. I always use Malt Extract Agar. To store a grown plate, culture, isolate, etc., they will be good in the fridge for years. In the end it all works for storage, just if you want growth you want it out in room temp.

About 5 days after my first agar attempt. 2 plates show signs of growth so far. Do these look normal?

Let them go a few more days to see how growth does. Top picture looks like contamination, not very full or white for mycellium. Bottom tray is to small of a spot for me to tell. Great thing with agar, even if that is contams, wait for healthy growth and do a transfer. You dont have to toss it and lose it all.

Looks more of a mold to me.
Healthy myc will be nice solid white

But its an agar dish, let it keep growing, if any healthy myc forms you can carefully transfer it
