Beginning again

Hello friends
Trichoderma has found it's way into my life :facepalm3: I've identified the error and am learning a valuable lesson about hidden contaminants. I was hoping for some advice on moving forward.

I'm going to take the plunge into agar soon, and was wondering if I should begin via spores to a plate, or begin with cloning a fruit from spores to BRF. Any experienced words of wisdom or pointers on which option is best would be much appreciated

Either or both would work so long as you take transfers and all that jazz. I took my first clone like yesterday morning, but I have done like 60+ plates including spores, LI, and grain. Just gotta dive in, but always look before you jump in and use common sense. Don't panic.

When it no longer exhibits irregular growth. Bacterial growth it pretty obvious, and so long as you let the myc grow enough from the transfer then take the next sample(s) from the leading edge of the growth you should be able to observe the differences and have a clean culture after two transfers. This us assuming your clean procedures are adequate. This guide should prove useful.

Would transferring a piece of myc from a shoebox that is likely doomed an option?

No do not transfer mycelium from grows into agar! Take a bit of spores or a drop of a spore syringe and put it to agar.

Spores are on the agar! So just to make sure, my goal is to observe the streaked plates, select a clean vigorous patch of myc, and transfer a piece of that to other plates until no contaminants are present. Once accomplished, I'll most likely have a polyculture, which can be isolated into a monoculture if desired. As long as my logic is proper thus far, my only question is: should I attempt to create an isolate? I know they're good for even canopies and consistency, however I read somewhere that polycultures may perform better. Is this true?

Monoculture = uniform growth. isolate or polystrain.
Isolate = a single strain that until tested could be weak, slow or even non fruiting

Most will recommend going for a mono culture from multispore (clean and uniform) then fruit and take several clones either from pins directly to agar or inner stem tissue clones from mature fruit.

That ensures the culture will fruit but not its potency.  Clones bring even more uniform results.
