Help with agar work.
    #25073032 - 03/18/18 12:31 PM (6 years, 11 days ago)

Hey guys. I’ve been reading about agar for the last couple of days fretting about doing transfers this weekend. My store bought shiitake clone exploded with mold. My grain to agar dishes seem contaminated and my Lionsmane clones look like the are contaminated now.

AA+ grain to Agar

Lions Mane

See the specks off of the LM? Are the AA+ worth transferring?

Any input is appreciated.

Would need better pics, I don't even see germination in the AA plate.

What's your agar recipe look like? Also you should try using pasty plates or just half pint wide mouth jars with the plastic pp5 lids un nodded or modded for sfd's

I use 1tsp (5ml) agar, 2tsp (10ml) potato flakes, half cup (120ml) of water, two drops of Karo, and a few drops of food coloring.  I’ll try to get some better pictures but these jars are hard to photograph.

You might want to check out FastFred's Media Cookbook.  You can download the file from the first post on this site.

https://www.shroomery.org/forums/showflat.php/Number/2996232/fpart/1/vc/1

Quote:

Potato Dextrose Agar (PDA)(FDA M127)
------------------------------------
200 g Potato infusion [dilute to 1L total, ~4g solids]
20 g Dextrose
20 g Agar
  1 L Distilled water (dH2O)

To prepare potato infusion, boil 200 g scrubbed, sliced(unpeeled) potatoes in 1 liter distilled water for 30 min. Filter through cheesecloth, saving effluent, which is potato infusion (or use commercial dehydrated form). Mix in other ingredients and boil to dissolve. [Dilute to obtain 1 L final volume] Autoclave 15 min at 121°C. Dispense 20-25 ml portions into sterile 15 x 100 mm petri dishes. Final pH, 5.6 ± 0.2.

Medium should not be re-melted more than once. Medium powder is available commercially but may require supplementing with extra agar to a final concentration of 20 g/liter. To BBL or Difco dehydrated medium, add 5 g of agar.

The broth is clear to slightly opalescent and yellowish in color. [1][2]

You're missing some type of sugar in your recipe.  I don't feel like trying to convert the weights to tsp and TBS and the volume to cups but I'd try adding a small amount (maybe 1 tsp) of Karo corn syrup, honey, brown sugar, or even regular table sugar next time and see if that makes a difference.

I'd also suggest getting a cheap scale and some way to measure volume in milliliters if you can.  I fought the metric system for years but it really does make this sort of work much easier.  You can get by without it but it is much easier to scale up or down a recipe if you just have to move a decimal point.

I’m sorry I have been putting a few drops of Karo in it. The recipe is from pasty’s plates. I’ll fix the recipe in the earlier post.

As far as the other jars are concerned it's really hard to see what's going on in there.

I’m pretty sure they are all going to be trashed.

Dude those plates look really good. It looks like your first transfer bounced around a little and the myc grew where it rubbed off.

Wish I had a cleared picture of the first pic.
