Pasty agar plates, how does this mycelium look?
    #25034135 - 03/02/18 05:11 PM (6 years, 27 days ago)

If you can see it well enough, how does it look, I have 6 more like this that all seem to look pretty similar, I tried to create a little divot with the needle when i did these 6 or so days ago but sometimes I would miss a little and the drop spread out across the surface. Either way they have been sitting in a relatively cold room for the most part so im sure the growth has been on the slow side. Can anyone tell me if these look clean or relatively clean from that image?

I know my sterilization process is on point cause I let all the plates sit for 2 weeks after I took them out of the PC and they where completely clean still after 2 weeks. As far as I can tell these look okay, but im no expert , this is my first time working with agar.

Not gonna lie dude that looks like snot. Harder to tell from that picture but to me it looks like an off color slimy blob.

Do any of them look white to you with threads?

yea, ill take a bunch more pictures give me a sec, and you guys could be absolutely right. But tbh when i shine the light through the top of the plate it looks totally white when i shine it through the side or the bottom it looks kind of yellow which could be due to the fact that its reflecting off the green agar (i put green dye in the agar)

If anything colored agar should make the white stand out more but lets look at your pictures.

Here is another one, as close as i could get it. Also i think it was the flash that was making it look yellow, no flash this time.

that looks a lot blurrier than when i took it on my phone, hold on. ill grab a few more

You said you had 6 plates? Hmmm... that one looks a little better but still hard to make out.

Can you open one up and get us a shot from above? If we can confirm ots healthy mycelium then thats better than losing all plates.

Sacrifice one to save many.

But i would prefer to see the first one opened

I think it's good, the thick white being multiculture and the lighter white being r culture. If you have more plates and work in a SAB then open one up and do some transfers, you should be able to tell what's up without the lid.

I do have a SAB and thats the plan. Would i take samples from the center of the culture? Or doesnt really matter. Also will be first time doing agar to agar transfers.

How many transfers should I go for per plate? 1 plate to how many?

My guesses so far:

1st plate all bacteria.
2nd plate some stranded bacteria-ridden mycelium in the center surrounded by bacteria.

I'd start over. Or if you must, transfer a scrape from just off center. GL

Hmm, this is my 2nd time doing agar and the first 2 plates where obviously contamd by my eyes(it was all the same batch of agar I just used the first 2 plates quickly and let the other 6 sit for a few weeks to see if my sterilization was on point), this time im having a bit of a harder time relenting. I agree that the first pic may be contam but the rest of them (ive only taken pictures of 3 of the 6) dont look all that bad to me, im not saying they dont have any contam, but I felt like I was extra careful this time in the sab and I know for a fact that it wasnt my sterilization process this time because I waited 2 weeks to make sure nothing grew on the agar and nothing did.

You could absolutely be right, and I bet if I open up the first one and take a picture it very well could just be straight contam. Maybe ill just sacrifice that one and open it up now so you guys can have a definitive answer on what it looks like and go from there. Even if that one is just straight contam though im not convinced the other ones are yet.

Also not sure if this would make any difference but the agar I made for this batch was probably on the stiffer side, not sure how much that changes how the mycelium looks/grows.

Opened up the suspect one, this is what it looks like.

Didnt smell like anything really.

My flash seems to have a yellowish tinge as the second pic definitely looks more yellow and that was with flash, first one without flash.
