First Grow Log - Agar, Oats, Monotub. 4/12/18 - PINS! 1

Made some agar plates using Bod's Agar Tek. Put one drop from a B+ MS syringe in the middle of each plate. Let it grow for a couple days took some pictures at the first sign of growth -

They looked pretty good so I let them grow a bit more then took samples from the best ones and transferred to more plates. 7 days after the first transfer I did another transfer. Here is what the second generation plates looked like -

Note that I had about 20 plates in each "dish generation", I'm just posting some of the better ones here. I never saw any blatant signs of contamination on the plates, so hopefully I'm doing something right with my aseptic technique.
With the 3rd generation of plates growing strong, I started to get my spawn jars ready using Bod's Oat Prep Tek. Got a 50lb bag of oats at Tractor Supply and followed the tek to a T.

Before PC

After PC

PC'd for 2 hours at 15 PSI. Now it was on to the agar to grain transfer. I took multiple (4-8) chunks of the outside portion of mycelieum on the agar and transferred them to a single quart jar. I wanted a quick colonization and I had a ton of plates, so I used a whole 100mm petri dish for most of the jars.

I may have let the plates grow a day or two to long, as it almost reached the edge of the plates, but I think I should be ok. 7 jars complete right now, and I just took the pressure cooker off the stove for 7 more jars. Should be enough for two unmodified monotubs (Bod's tek once again).
Is it ok if I keep the jars on a shelf in my closet or do they need light to colonize?

They will do just fine in your closet for colonization. Keep up the good work.

A little less than 5 days since I did agar to grain on 14 quart jars now.
Here's what they were like after three days:

And here is what the looked like a couple of minutes ago:

Note that I didn't shake them after I transferred the agar wedges in. They were kind of just sitting on top of the oats and the myc was taking a while to reach the oats and colonize, so I shook half of them up as an experiment. I read some posts that it may be a good idea to shake once the mycelieum reaches some of the grains, so those grains get spread through the jar and become multiple inoculation points. I also read some things saying definitely don't shake until 30%-50% colonization, hence why I only shook half.
Here are some shaken and unshaken jars side by side. Will update once I start seeing growth in the shaken jars. If I did it correctly I expect the shaken jars to recover in about 48 hours and reach full colonization a bit quicker than the unshaken jars (which I will shake at about 30%).

Let me know what you guys think.

Personally, I'd probably shake them to distribute the myc. Your grow looks great so far.

Do you think it'd be a risk to shake the rest of them? I wasn't sure if they were grown off the agar enough, which is why I only shook half of them.

I don't think its a risk no. I would at least shake the jar so the agar is covered by the grain. It'll probably be fine shaken or not though

Cool cool, I just shook them all up. Hopefully I see some nice growth in a couple days. Thanks for the advice!

Here's what they look like 48 hours after shaking. The mycelium went from invisible after shaking, then little dots that were hard to see at 24 hours, then lots of growth in the past 24 hours to get to this. I'm thinking they may be fully colonized a week from now, which would put it right at two weeks from colonization

Question - should I shake again at 50% or should I not bother since I already shook once the mycelium transferred to some grains?
I plan on saving one of these 14 jars as a grain master for future runs. What the best way to store a grain master jar for semi-longterm (a couple weeks)? Fridge? Or can I just leave it in my closet?

Wait and see at 50%. You probably won't need to that soon.
Fridge for a couple weeks is fine. Make sure it's fully colonized before fridge

So it's been 13 days since I inoculated with agar wedges, and 8 days after I shook all the jars.
Here's what they looked like 5 days ago:

And here's what they look like tonight:

I think they look pretty good and I'll hold off on shaking again because the growth is pretty even throughout the jars.

Thanks man, it's good to hear all these words of encouragement, especially after my first attempt last year was a complete failure hahaha. I browsed this place for months to get more info and this grow actually looks like it will succeed. I made sure to have enough jars for two monotubs just to be safe though hahaha
