Agar Supplies and Questions

Hi all, I'm about to start experimenting with agar soon and I just have a couple questions before I begin

Which is better for agar work?
125ml (4oz, 1/4pint) regular mouth mason jars or 90x15mm petri-dish?
Personally, I feel like petri dishes are better for agar work since they're flat and has more room. Whereas jars, especially with regular mouth is quite deep compared to petridishes and would require tools to be manuvered over it, which could be a source of contamination. What do you guys think?

As of right now, I don't have scalpels, but I do have a rusted general xacto knife on hand. Would that work or should I get new blades? What would constitute as a cutting tool that's easy to sterilize? Butter knife perhaps?

I also figured that I would need tweezers to pick up pieces of agar later for transfer. I do have a pair of plastic coated tweezers, would those work? Or syringe tips ok?

Lastly, for spore prints onto agar. What is the best way to inoculate agar with prints? Should I simply scrape off some spores onto the agar or should I make syringes?

I know this is a lot of questions, I just want to hear from you experts some of your experiences working with agar so I don't make the same mistakes.

My Agar Recipe:
20g agar-agar
30g Ovaltine
1L Water

ovaltine agar? interesting. i've never tried it. i use Light Malt Extract and it works wonderfully.

i like petris wrapped in glad cling wrap the best.

Anything that you can flame sterilize will work but you'll definitely want a scalpel if you do much of this.

i used to use tweezers for grabbing agar pins but now a days i do almost everything with my scalpel. it's rather easy to pick up a wedge with a scaplel once you get the hang of it.

you can make syringes or scrape spores off. just remember that they are tiny tiny tiny little fuckers and you don't need many.

Thanks for the reply, I saw the ovaltine agar recipe in one of the threads. Couldn't find it right now but I think it'll work since Ovaltine's main ingredient is barley and malt extract. It does contain salt and magnesium carbonate but I figured that in small amounts it'll only make the mycellium stronger :laugh: Will let you know how it goes

Not saying it won't work but why not use LME or brf? Even dog food has more merit then ovaltine in my opinion.

Also are you planning on pouring like 50+ plates cause I don't even use half your recipe and I always have lots left over!

And as far as containers go I've actually been prefering my homemade sfd/ship wide mouth half pints over pouring petris or dealing with pasty plates and having to use napkins/paper towels and aluminum foil. My mason jars will last forever and I can pour my agar in a bunch, food color each one whatever color I want if I'm doing large batches of multiple varieties and it makes sorting and labeling a breeze.

I've tried it all so far and half pints and the smaller jars are my go to's for agar work now.

well this is my first time experimenting with agar so I don't know what's the amount required for plates and jars. I figured Id be safe and make extra, but I'll probably cut the recipe in half now that you mentioned it. I don't want to use brf since I want to start cloning and there's no way for isolation and cloning work using brf.

As with ovaltine, it's the only malty thing I have on hand atm. I figure I'd put it to use since the main ingredient to ovaltine is BARLEY AND MALT EXTRACT. I currently have 1/4 pint jars regular mouth. I couldnt find the widemouth option anywhere where I live.

I'll look into dog food, but if worse comes to worst, I'll probably just buy some malt extract from a beer store or something (if ovaltine doesn't work out)

U can use dog food at 2% ratio. 2% dog food 2% agar 96% water. Ive never done it myself but everyone that has raves about it.

why not use PDA ,
ive got 2 sets running at the minute 10PDA and 10LME and have noticed that the PDA are massively outgrowing the LME , i know this is going against the grain, maybe ive miss calculated my Light malt dosage and either made it to strong or weak but the PDA have spread out in a week.

im using Instant Mash and its a doddle!

Ive excluded yeast but my next batch ill be adding yeast

I wouldnt use yeast... and people usually have dog or cat food already available and it works great from what ive seen.
