How do I store my inoculated agar plates?

I searched and could not find an answer to this. The agar TEK I have seen says to store pure agar with saran wrap or something covering the sides, preventing air transfer. Is this true for storing inoculated dishes?

absolutely. you can get away with not wrapping your plate but it is not worth it in my opinion. you have two options, parafilm or glad cling wrap. supposedly saran wrap doesn't breathe and glad cling does. I've never tried saran so i can't say but i've done a ton with para film and a ton with glad. you can see them here
glad cling wrap is my favorite. just by a roll and cut it into pieces that are wide enough to cover the edge of the petri. i clean it off with iso and then discard a couple of feet. i like to wrap it around twice.

Always wrap dishes 360o around asap. I've never used saran wrap but I would assume it works just as well as the press-n-seal that I'm partial to. Nasties can weasel their way in through places you wouldn't expect.

I use parafilm and usually make it around 1 1/2 times with one piece. I use 2 because after awhile parafilm dries out and the edges of the plate will rip the parafilm (for me at least). Using 2 pieces stopped this.
As far as inoculated plates, freshly inoc'ed just leave em sittin out somewhere to colonize. If they are colonized and you want to store it for latest use put it in a ziplock and put it in the fridge. For extra protection I put them in a lunchbox with ice packs (not frozen) around them for an additional thermal barrier since temps swing in a fridge from repetitive opening.

That's a good idea for long storage thanks for the tip brother.

Correct. Parafilm has a moisture content.

Thank you! I was just about to do my inoculation and wanted to make sure I wasn't going to suffocate my spores or something if I wrapped them.

It won't, plastic wrap can breath a little and parafilm is made to breath to allow GE.

This dish is wrapped in Saran Wrap and is in an opened ziplock bag. Seems like it works to me.

For about how long would they be good / viable in the fridge once fully colonized?

A good while. I xfer mine twice a year, with no notable loss of vigor. And there's numerous accounts of much longer than that. For longer term storage (2+ years) slants are the way to go.

I've read that they can be kept for up to a year before needing to be transferred to new media, although in most circumstances the agar will dry out before then.
I haven't kept a culture in cold storage for longer than 2 months, but I'd say 4-6 months
If you really want to keep a culture around for awhile you need to look into slants, but you'd really only want to do that with badass clones and isolates. No sense in keeping something around that long if it isn't primo.

What about agar plates that aren’t inoculated with anything? Can you store those in the fridge as well? Or would they be better stored at normal room temperature. If they can be stored, how long would they be good for? Sorry if somebody already covered this, I got a little confused reading some of it. My brain hurts.

I store mine in a ziplock, no parafilm, in the cabinet with the rest of my myco supplies. It keeps my sterile tek in check too.
If you plan on keeping them around awhile, stick them in the fridge.

Ok nice. I keep my syringes in the top of my fridge door that has a cover on it, sounds like a good spot for agar. Nice, thanks!

Yeah that would be a little easier to get away with than not wrapping a petri. My clumsy ass would pull lids up on my stack of colonizing plates if I didn't wrap em lol

Until they dry out, depending on your house and how you store them, how stiff your agar is, and how thick you pour that could be weeks or months.
Wrapped plates retain their moisture better, but I don't keep prepped plates around for more than a couple weeks and I don't feel like wrapping and unwrapping more than I need to.
