So I've been reading/learning here for the past few months and slowly ordering and buying supplies/materials here and there. At this point I pretty much have everything I'd need except maybe a few additives/nutes and a flow-hood. I'll be Starting from MS from a vendor. Its hard to get an outlined game-plan. Soon as I have a game-plan and think I'm going to do things "this way", I run into issues and learn It's better to do it "that way". Originally The plan was to go from MS to LC, but read that was disastrous and potentially a great waste of time.
Next was pointed towards going towards the direction of going MS>Grain>Agar.By that time the cultures have had time to battle it out and would make it easier, since Spores like to germinate and prosper on grain more than on agar.Now I'm reading that going MS>Grain is also not suggested?
I have almost everything at my disposal at this point, save a flow-hood. Which route should i travel?

Spores>agar>grain seems to be the way to go.

Germinate on agar, 2 or 3 transfers to confirm cleanliness, inoculate grain or LC

Spores to agar. Then 2 or 3 agar to agar transfers. Then agar to grain. Then spawn to bulk substrate.
Flowhood is far from necessary. Nutes/additives are known to be useless at best and detrimental at worst

I first started like this. Spore syringe to BRF cakes then clone/print to agar then grain.
I never did spore syringe to grain or spore syringe to agar. Dry spores (spore print) to agar yes !
At the moment you dont need a flowhood or anything fancy. SAB for the win !
Make lots of backups, agar is your friend.

Tons of threads I come across are very old and sometimes its hard to put these in order and sort through the most recent teks. I have activated charcoal (from hardwood). Is adding activated carbon to agar still beneficial, or is that just for better view of myc on agar? I'm still about 5 days out from MS arriving. Should i pour my agar yet?

I would wait till u get ur spores b4 pouring agar. But im sure u could pour it now if u wanted to. But dont get in the habit of prepping things that many days in advance.

You shouldn't have any need for charcoal. And i don't see what it would hurt to go ahead and pour your plates now if you'd like. Plus it'll give you a chance to potentially see contams pop up in case your pour technique needs improvement

True.
I myself pour agar days in advance, to discard eventually contaminated plates.

Make yourself a nice clear tote with two armholes in it thats tall enough to pour one quart jar directly into another. Buy or build a flow hood if you decide you need one after you get sterile tek down in a box. Germ the spores on agar do a couple transfers until its nice and clean, put half on grains and wait for the other half to pin and clone that bitch to agar or if your sterile twk is on point straight to LC. By the tine the clone is on grain youβll be just about done with the first flush from the A2G.

U never go froM MS to grain because of two main reasons:
1) Possibility of contamination from the syringe/spore print.
2) No control of the genetics.
I did it anyway with no issues, but its smarter to use agar. Its lazy and risky to do MS to grain.
