What is the best way to apply spores to agar?

Hello friends!!!!! :mushroom2::grin::thumbup:

      I recently acquired some P. Galindoi spores and would like I to get them going on agar so I was just wondering what is the most efficient way to germinate them on agar??  Should I make a spore syringe or should I apply the spores straight to the agar using an inoculation loop or something??  I will admit I am relatively new to agar and have only performed tissue transfers to agar, never spores. 

      After using the search function I found a lot of different info, some recommending making an lc, or a spore syringe..... But I would like to know from a trusted source what the best method is for applying spores to agar if ya could please?? :grin:  I will be using Pasty plates. (Thanks Pastywhyte!!!)

        This may seem like a nube-like question, but I guess am somewhat nube-ish when it comes to the Straights of Agar!!*

          (*Mushboy coined phrase :wink:)..... Also what grain substrate is best for a truffle producing variety like p. Galindoi??

Use a loop.  The stones in my sig were done on rgs, but I've used wheat and rye berries as well.

Apply spores straight to the agar using an inoculation loop.  Dab one spot directly in the center or the media, or streak the plate with the spore loaded loop.

when i started out i just used the damn flamed scalpel. got tiny speak of spore and sliced through the agar.



keep it simple at first. you dont need an inoculation loop. ive never used one to germ spores:shrug:

Ahh, that's good because I don't have one!!!  Haha!!  :dancer:

    How many plates would you guys do up for the first wave...?  Four plates to transfer from should be good ya think?

Four plates will be fine. 

Although swabbing spores with a blade will work the sharp edges damage spores. 

An inoculation loop is designed with a smooth edged loop at the end that does not damage spores among transferring.  With an inoculation loop the option of streaking the spores onto a plate is had also.  Streaking allows more germination points thus giving the cultivator more options of transferable culture, as well as, more areas to transfer clean myce away from contaminates so would they happen to break out.

Thank you ham-man!!  Should I just sterilize some qtips in my PC and use them to pick up some spores and streak the plates with it?  Would this work similarly to a loop?  :cheers:

I always use swabs. Very forgiving. Some are super easy to open in a SAB. As long as you don't get your hands over your work you'll be fine. I like using my noc loops for choosing which mycelium to pull. Never thought of them for spores seems kind of... more difficult than it has to be.
