An experienced grower, PussyFart, did an experiment **link** where he used 10 PE isolates from good PE fruits and grew them out with widely varying results with only 6/10 tubs producing fruit. I think the lesson to be learned was to not find an isolate for the sake of finding an isolate (especially if you're going from MS straight to agar - you'll have no idea what you're isolating), but to find an isolate once you know you have a good set of genes that produce quality fruits.
Strain isolation from culture started by spore really isn't that hard though. You keep transferring the culture until you can see obvious sectoring. Then take a transfer from each sector to a new plate. Those should be isolates, sometimes they need another transfer if you grabbed a transfer from a couple sectors by mistake. Now you have 10-20 isolates, which you label and test. I use substrate bottles, each bottle gets inoculated with a different isolate , I fruit them in the same monotub to fruit them side by side. So a bunch will do shitty, some might not fruit at all. But there could be a real winner in them
Its really pretty great. But for our purposes, I think cloning might be better suited

Idk...i mean look at the struggles pasty has been through with his rustywhyte, cloning can be and often is just as much of a headache as isolating Either way...put your work in
