Happy Friday to you folks!!!

      Hope your all havin' a good one!  It's snowing where I'm at!!! Haha..... Quick question for you Knowledgeable Souls.  When would be the "best" time to clone this pin:



    Its the big one right in the center of the pic.... If came up first and is the healthiest one of the bunch.  Nice shape too!

      My problem is I would like to be able to let it mature and print it, but I also want to make sure that the tissue I clone is still vigorous enough to colonize my agar.  So if I wait until the fruit is mature and print it will it's tissue still colonize well even though it was from a mature fruit??

Remove the cap, and Clone it when it's mature by taking an inner tissue sample of the stipe and placing it on agar.  The cap needs removed so no chance of spores get onto the agar and germinate, then print the cap.

Thank You Bodhisatta-San!. Just for general info's sake.... Do you have a preference for when you harvest clone specimens?  Do you usually print them as well or use a technique similar to what is described by hamloaf?

Thank You hamloaf-San!!! . That sounds awesome!!!  I should transfer the inner stipe tissue DIRECTLY to the agar correct?  I was thinking I could put it into a Ziploc baggie for a couple days first and let it recover in the bag before adding it to agar??  Or is that just if using part of an immature pin?  Reason I am asking is because I have seen it said where some people will do the bag thing???  I am just looking for the best techniques! :rockon:.... Would you recommend that or putting directly to agar???

        Basic question, but.....:shrug: lol.

Plenty of others to print...harvest before the veil tares and clone it, no need for recovery in a baggie that leads down bad roads..strait to agar.

What fruit you print from doesn't really matter since you're working with a stable variety so you may as well print any fruit that isn't retarded. So clone the cool one and worry about prints from the others like cron said.

I clone any time before its mature if possible. Because why not its technically better to do so as well. But its not like an overly mature fruit just doesn't work or sucks for cloning. We always heard shit talked on cloning grocery store mushrooms. And even that usually works well.

Better is subjective.  If searching for desired traits it's more ideal to let the fruits mature first, then clone. 

TAG89, do not put fruits to be cloned inside of plastic bag for a few days.  They will water log, breed bacteria and ruin the tissue sample.  Correct, put the sample directly to agar in sterile conditions, and be ready to do such immediately before harvesting the subject.

No, it really is better to do it before maturity because after spores drop apoptosis starts. Hence fruits falling over.

Nah, that's irrelevant.  Take the clone before the fruit sporulates in order to avoid cell apoptosis.  Also, remove the cap before placing the subject into sterile conditions so no spores get onto the agar and germinate.  Again, if hunting for phenotypes, gather mature fruits for cloning otherwise you might end up with trays full of short and stubbies as mentioned up thread.

Ok.... I'm sure there would be a window of time to snag the fruit AFTER it has matured but BEFORE the spores drop correct...?  You could keep it growing long enough to see it's characteristics, but then snag it it after the cap flattens (mostly) but before spores drop..... In some cases you can harvest the cap before spores drop, and then they will drop during printing as long as you pick said fruit at the proper time right?

    So one could harvest a fruit during all these "perfect storm" conditions then probably right?  You'd have to be watchin' em like a hawk tho!! Lol

I like to clone from pin clusters a lot. I slice out one of the pins for cloning and then keep an eye on it to see how it turns out. Best of both worlds! :lol:

Again, not really, because in that case you could end up with a culture that produces stubbies as mentioned up thread.  Want substrates that produce large fruits very quickly?  Let fruits mature and grab an inner tissue sample of the fastest growing, largest fruit(s).

I was under the impression when cloning from clusters it doesn't matter so much which part you take your clone from?
I figured if I took a pin from a cluster I can then watch how the rest of the cluster turns out
Honestly Ive always been confused about clusters.

I don't always get clusters when I clone from clusters but I usually do

I always thought it was the opposite.... In a cluster, each mushroom still has it's own individual genomes.  When you pick from a cluster for cloning it should be the biggest, healthiest and fastest to maturity I believe.  :thumbup:.... I'm no expert but pretty sure :smile:
