I got 12 jars going with some pre-made jars/spore that I haven't tried before. Should be exactly a quart of grain per each jar. I tried to order the pictures from worst progress to best; I have a wide array of results. 4 of them are pretty fully colonized and white, 3 of them are partially colonized and white, 3 are partially colonized and yellowish, and 2 are simply wet with no sign of mycelium. I plan to use the 4 best to start a monotub soon, but what to do with the rest of them?

They have been in temperatures of around 70 degrees the whole time and have been colonizing for something like a month or so. Are the yellow ones done? The wet ones? I have some spores sitting in a syringe but figure its not really gonna help at this point. Any advice as to how to cope with the lackluster performers would be much appreciated.

The last pic is the only jar you posted that is done. Also. Temps in 70s are perfect

They need to be mostly white like that. Theres another jar that looks close but i dont think 100%. A bunch look clean enough for a project tho, so, go you! :smile:

I would make them yourself tho. Much cheaper, and youre not gonna wanna keep ordering pre-sterilized jars. Strangely in my early days tho when i used pre-sterilized, it usually worked out well. Its just the $ and learning to DIY

Buy a bottle of malt extract agar mix. And 25lb of rye to start. Some coir. A pressure canner. And Synthetic Filter Discs. Gloves and alcohol

Then youre ready to go and no need to order more materials (besides maybe syringes etc)

Next time, do agar first before doing QUART jars..

START ALL OVER AGAIN.

Its smart to use agar with "quart" jars but shit, all the times i didnt and it was fine.

I dunno if vendor syringes are dirtier now or what, but you used to be able to just knock it up no problem. 8/8 jars would grow well

Spore syringes are almost never totally clean due to the nature of how they are made.
Prints taken from indoor fruits are generally much cleaner than outdoor specimens but unless they were grown invitro and taken under sterile conditions it will not be totally clean. 

When we grow from print, we have to swipe it to agar and that gives us a chance to clean it up. Spore syringes skip that step so whatever contams were on the print get into the syringe . Its why I usually have to transfer away from some bacteria if I inoculate agar with spore solution.

BRF cakes are quite forgiving and a decent spore syringe will give me a near 100% success rate most of the time.
But grains? Not so much. Doesn't mean there is anything wrong going spore syringe to grain but sooner or later you are bound to run into some trouble. Could mean losing an entire spawn run!

^ive had many successful MS (syringe-to-jar) grain projects but the last batch of syringes i got, its just contamming everything, and i guess agar is the only way to avoid this annoyance. I JUST knocked up some plates with Maz, and hoping for the best. I grew out this same maz on grains no problem. Do vendors still return syringes if theyre not clean after 30 days? Cause that used to be the deal. They would be clean enough for big ass mushroom bags even. I wonder if vendors are noticing that more people are using agar from the beginning, so maybe less effort in keeping the syringes clean for MS??

i have no idea what the truth is, maybe gotta test some more syringes. This was one batch from one sponsor.

I dont mind some agar work, but i liked how places would guarantee clean syringes, or at least clean enough. (For microscopy, of course)

Put spore solution on agar and if u get the same problem on agar its DEFINATLY the syringe

It is, i tried it lol. Agar turned the sams color as the jars. Blue as fuck. Fast. All plates at once lol.

So yeah. Trying some new syringes with some older but clean-looking refridgerated agar.These are maz. If that doesnt work out, i have a new batch of plates ready and a newer syringe. I kinda just wanna see if this syringe will do the same thing or not (same source as the fucked up syringe, and i have some brand new ones from somewhere else)

I hope it doesnt take more than 1-2 diff syringes before i get something clean enough
