Imo it depends on whether you leave the bags in PC to cool overnight. If so, air sucked in by (de)compression should be sterile (and humid btw).
I just use pieces of tyvek envelope but i might wanna do something about that.

Maybe EZ felt is great here too esp if you don't let it dip in substrate.

There are some members that don't use tyvek when sterilizing their bags although I still do. I'll probably ditch it soon though

Just wondering if I had a colonized sub bag, could I expand that bag into more bags? The bag is supplemented hardwood sawdust

You could have if you had already set it up to be used as spawn.
As in already assembled into a spawn bag "aseptically".

You can also try wood chips + whole grain flour.

I had issues with wild Tremetes Versicolor clones. On agar they contaminate very fast. Had some growth, but cleaning is hard and i lost couple of dishes.

1.

In SAB I cut piece of tissue, wash in H2O2 and then in sterile water. Put on agar.
High contamination rate, very diverse. Fail.

2. Wash bleach solution, wash sterile water. Put on agar. Still contaminated. Molds for most part. I dont like working with tissue/mycelium that molded in dish.

3. Decided to use toothpicks. Wash in H2O2. Then bleach solution. Then sterile water.
repeated few times. Put piece of tissue on agar between small pegs.
Put on just sterile toothpicks.
Got nice growth. Transfer.

This I can work with.

Unfortunately my dslr is broken, so no pictures, but my jars of reishi on WBS seems to be starting the fruiting process. The mouths of the jars are about 2/3 full of uneven mycellium, which has taken on some color, light tan to light brown. The mycellium within the jars has been producing irregular colored areas as well (brown), and one of the jars (the more fully colonized one) has some of the characteristic amber reishi metabolites in a pretty cohesive teardrop under the rim. Both have a couple of distinct brown bumps on the top about the size of a dime in diameter, slowly rising up, which I assume is the beginning of antlers. The more colonized jar also has some darker colored mycellium creeping over the side of the mouth making contact with the bag, trying to get down to the air I suspect (as the bags aren't sealed, just zipped tight, there are tiny gaps on each side).

I've had them sitting with the tops off, covered by plastic bags in a ~60-78* room, getting moderate amounts of sunlight.

As fun as these are to watch I am itching to do some proper invitro bag grows so I can make some tincture for my grandmother. When working with bags is liquid inoculation the only way to go, or could I feasibly inoculate bags of mixed WBS/Coir from colonized agar in my SAB? If the risk is too great I guess I'd better start learning how to make LC, as even at $5/12ml that would add up pretty quickly...

How long should I pc lme LC? Can't find the sterization time...just need to know before I throw these jars in the presto..is 30 minutes long enough?

30 minutes is good, just don't do more than 45 or you risk caramelizing the sugars. I normally do 20@15psi with no problems.

Every time I cook a bag of grain in my pressure cooker it comes out soaking wet and then ALWAYS gets bacterial even when using gloves and a SAB ect. I have almost perfect success with jars. Should I just wait till I get a flow hood to do bags? I really need to increase my spawn production and pint jars are getting tedious.

Couple thoughts are to ensure the grain is bone dry on the outside before loading and if it’s still very wet when it comes out, you might let it sit a day or so before inoculating to allow the moisture to be reabsorbed by the grain.  A hood is a must for clean/practical bag production.

This year will be my first attempt at going "large scale". I have been selling off oysters to family and friends whenever I have them but would like to get regular production going.

In anticipation for the growing season (in my cold garage) I have about 50 agar plates going. I plan to make 40 grain jars each weekend to meet my quota. I go straight from petri dish to grain then to substrate for fruiting. Would it be more efficient to expand my grain a bit and store it in the fridge till its ready to go into substrate for fruiting? My main issue so far is keeping up a schedule of making spawn and bags for fruiting. Im thinking a monthly large batch of grain would work better?

edit: just found my answer using search

@quad my pressure cooker is tiny haha. Ive been spending all my spare pocket change on growing supplies. Hopefully by the end of the summer Ill be able to get myself and A.A. cooker and flow hood if I dont fuck up :cool:
