Harvesting questions

So one half of my tub had a nice flush, while the other half is just now forming pins.



-Would you harvest all of the mature fruit and pins together to start with a clean slate? Or leave the pins on the slow side to reach maturity?

-If I pick the mature fruit instead of cutting at the base, will there be no chance of a second flush at that location?

These particular fruits are so thick and close together with fatty stems that I don't see how I'll be able to cut them easily. Ill have to take my sweet ass time if it's absolutely necessary though.

-I would like to take some spore prints and pins for cloning, is it best to do this in a SAB?

I think my SAB is just barely large enough to work.
I tried taking a clone without a SAB and the agar filled up with contams so fast the mycelium didn't have a chance to establish.

Last question:
-Why do you think the pinset is so uneven?
If it were random patches I would assume uneven colonization, but because it is bisected I can only assume it is environmental. Perhaps my polyfill are stuffed too tight on the slower side, thus restricting FAE?

Harvest the mature fruits yea.

You have to use a still air box for agar work, never use agar in open air.
If you're going to clone a pin, take the pin into your SAB on your scalpel and drop it onto your plate. Young pins will grow pretty vigorous myc and it will outrun contams , letting you get a transfer

If you are using a mature fruit , bring it into the SAB and tear it open with your hands, and drag a sterile scalpel along the virgin tissue to get a piece to drop on agar.

Yes, you want to pick the ones that are ready and leave the immature ones..... Definitely do printing and cloning work in an SAB. 

        I like to wipe a large rimmed cup out with iso 70% and place it over the top of the cap you are printing, while you have it in your SAB.  Just take your time harvesting.... Grab the ones that are ready by the base and lightly twist one way or the other (whichever is easier) and lightly pluck em away from their clusters and you can use a knife to assist with it....:thumbup:

How to take clean spore prints

Is your sub level? Did you mix your grain and sub evenly? That could cause it to be uneven in growth.

Whenever I’ve twist and pulled mushrooms, I still got mushrooms from those locations in later flushes. Cut them if you want, I don’t think it matters very much, you will get more mushrooms either way.

I did my best to mix evenly, and to make AS level as possible. It appears to still be fairly level.
I have noticed the sub has pulled away from the walls on the mature side a bit more than the slower side.

I'm not able to visibly see any spores being dropped on any fruit, but they all seem fairly mature...
I have heard that these mutant KSSS don't spore print well. Hopefully it's not a major issue, some of my previous flushes of the same genetics produced spores.

-Is it not a huge concern of how I transfer my fruits from the tub to the SAB?
None of the teks I've read discuss it, but I was thinking to sanitize a shoebox and then place them in that while inside the tub to transfer them to the SAB.

That’s strange. How did your agar look? How did the spawn jar recover from a shake? Maybe somethings up with the spawn.

Well this tub is actually spawned from MS that was transferred G2G three times before.
Also, I'm pretty sure the spawn was bacterial, but it seems the bacteria doesn't inhibit colonization because when I moved the mycelium to agar the bacteria was present but the myc totally ate the bacteria up with the agar.

I just started working with agar so I'm still working on getting a totally clean culture to further isolate.
My next jars I will be starting from agar.

No you can carry a mature fruit right to the SAB. By tearing the fruit open with your hands, (don't slice in, it will push in contams from the outside, just tear it) it exposes virgin mushroom tissue, which is sterile

I know sometimes people have a hard time printing that spaceship phenotype.
You may want to sterilize some cotton swabs and use those to collect spores.
