Have a few agar questions, all help is appreciated!

So i have been looking over pastys easy agar tek for a while now and im gonna give it a go. Would it be ok if i made the pda and then immediately poured the mixture into a glass bottle? Then maybe and hour or 2 later put the bottle along with my plastic petri dishes and alchohal in my sab and just pour the mixture? Never done agar before, it seems confusing for the first time. Thanks, also if you have a really easy way to make pda and pour that would be amazing!

Agar should be poured under as near sterile conditions as possible. Additional steps and prolonged exposure to air is likely going to result in nasties on your plates...

Thanks for your reply! So i couldn't do what i said even if i used a steralized bottle and a sab? And ofcourse poured in the SAB? what do you use?

If you're pouring plates, you want to mix your agar in a bottle, pressure cook it, 15psi for 30-40 minutes, wait for it to cool down take out and pour in your SAB if that's all you have.

If you only have a SAB, I'd look into doing no-pour agar plates.

Ok awesome, would you recommend just following pastywhites tek or do you have one? I only have a SAB but i was reading a pressure cooker will melt plastic dishes. Should i just order some glass ones?

Na man use some little jars I chuck 10g light mault extract with 10g agar into cold water bring to a simmer as stirring make sure it’s all dissolved into the water then I pour about 30ml to each jar and pc for 20mins.
I bang a hole in the lid and cover with tape to help with condensation and wrap tight with foil to stop water entering the jars

1 I mix my agar.

2 Put in glass container and microwave, CLOSELY watching until it reaches a boil. This is to make sure it is dissolved and properly mixed, I use MEA.

3 I then leave lid loose and cover with foil.

4 PC at 15 PSI for 30 min, NOT 90 that will fuck the agar up. Agar does not take the same time to be sterilized as a solid mass like grains.

5 Agar need to cool but NOT solidify.
I like to put my bottle in 140F water, with a thermometer. I watch it and start pouring at around 120-125f. You can handle the glass at this temp but it will start going solid quick. If pouring a lot of plates maybe start a little warmer. To warm will cause condensation.

I pour up to 80 plates at a time and start when the temp is 145 or 150

I did 10, I can see how you would need to start higher to get through all of them. Maybe I should start a but higher next time as I got some air bubbles trapped in them.

Yeah, i have to start a little hot to get it done before it starts to solidify. luckily, i invested in a huge ass flowhood so i can move pretty quickly and comfortably. I only notice condensation problems when i start above 150.

also, I have found that the cure to air bubble is to swirl the agar when i take it out of the pc and then let it settle until i pour it. I always get a few bubbles in my last few plates but they don't really bother me.

They were not terrible, and I was pouring in front of the flow hood, so I figured I let it get to close to solidify temp, and it just trapped them in there as it probably solidified in a couple seconds with the hood blowing on them.

This is correct, sorry, I was thinking grains. I PC my agar for 30-40mins. Though, if you leave it for longer, I don't forsee there being any issues?

What specifically does it do to the agar if you leave it in there for too long? Never tried.

Why are you telling other people to do stuff you have no clue on? From what I hear is it will cook agar but then again I have never pced for 90mins bc 20-30 is standard unless you are using grain water then 45mins

The sugars caramelize. Apart from that (conected though) the agar solution gets unnecessary dark

Water can steam out causing the agar to be stiffer than you want as well. 90 min would cause a fair deal of moisture loss in agar compared to 30-40 min.

You can pour in a Still Air Box, taller SABs are better.

It sounds like you want to start with no pour plates.
Pasty uses Glad Minirounds, they are PP5 plastic and can be sterilized in a PC without melting.

Pasty's is a good tek , I use them a lot when I need to save on plates.
