Would you transfer from these plates?

Hi. I've been away for a couple of weeks and this is what my plates look like. First time I succeed in getting germination. Would you transfer from these now? If so, from where?

What sort is that?

Kinda messy.  There may be wedge locations.

Hi, I would transfer from the very edge of the puffy looking mycelium. I personally think it's mycelium. It grew from where you streaked isn't it? That at least tells you it might be mycelium (and it probably will)

Funny threads growing though so soon after inoculation..

I wouldn't go for puffy fuzzy myc myself.  honestly that looks like a poor inoculation.

Yeah, it grows from where I did a z-streak and all three plates look the same. They are three weeks old. Isn't this aerial mycelium we're looking at?

I would do more plates.  Honestly, unless it's good spore (bred for conditions) I would run a lot more plates.

It looks a bit homely, wired weirdly and just sorta wrong.  But could be cleaned up.  What's the goal?

I would want to be totally in love with curiosity to run a culture.  I don't think interest in shrooms is at all a desperate measure.

Weird strands on there! I'm wondering if this is an azurescens or something like that... the strands remind me of another thread I saw earlier, where there were strands between colonies on wood chips.

I suppose that they could be cracks, but it seems directly related to the mycelial growth positions, so it would surprise me.

Looks not much like the cube mycelium I've been getting on first my germination plates or transfers, though I'm new at this.

None of the plates are cracked. The agar is really thin so the z-streak has cut through in some places. There are wrapped in parafilm. Print is supposed to be z-strain cube. I'll try to transfer and see what happens.

Myc looks stressed and running from bacteria, gl, I’d swab new plates and do a couple transfers from this edge.

Sterile swabs right meow

Take a qtip wrap it in foil. Put 15 of those in a 1/2 pint with a lid any lid. Pc that bitch for 30 minutes.

Use those qtips to swab a print lightly without your hand ever going directly over the print.

Swab it just enough to make contact in a simple Z pattern. Do not press hard to disturb the agar. Pour thicker plates.

Or do what i did the first time. Scrape off a bit of a spore print on a flame sterilized xacto knife after it cools down about 10 seconds is enough. You only have to scrape about the size of a freshly sharpened pencil tip. The motion is scrape then turn it facing up then open your agar plate with yiur left hand if you are right handed. Without putting your hand directly over the plate gently lodge that tiny sharpened pencil tip amount of spores into the agar in the dead center once not twice or three times. Once. The reason is visibility and identification purposes.

Once it germinates which can take a while up to 2 weeks or more start a new thread here where we can tell you where to take a transfer from before its too late.


You didnt say your agar prep. I'd recommend reading bods agar thread to see if you have that down pat if not do no pour agar a la pasty plates link ----in my sig . C10 has a good agar thread and stro has a good thread on cleaning dirty plates.

Thanks for all the friendly advice, I really appreciate it.

I've made these plates with no pour Tek in glass petris (PC:ed fΓΆr 20 minutes), soft MEA, inoculated with a DIY loop as per Bod's DIY loop TEK, from a freshly received print (supposedly quality spores print), in a SAB and left the plates for 3 weeks while I was away.

I'll make some new plates without food color. The food color could be the culprit. I'll then do some transfers as well as start some new plates from scratch. I'll update/ start a new thread when I have something to show.

I was under the impression that glass petris were not well suited to no-pour teks, due to the possibility of the agar boiling up as well as the increased likelihood of condensation (and thus contamination). Maybe this isn't a hard and fast rule though. Due to these conclusions from my reading, I switched to poured agar for this recent jump into glass petris.

Not to say that this is the problem, but it is a possibility.

Exactly the problem. Glass petris and pyrex are pour only. No wonder the glass cracked.

I actually found poured agar into petris to be easier than no pour, at least with the PP5 containers I used originally. It might be worth it for the OP to switch.

You can do no pours with both Pyrex and plastic petris, there are quite a few tc’s who have written about it. with glass petris you just need to pc them after the pour but bring the pressure up slowly to prevent overboil(it’s a good idea to raise the pressure slowly for a plethora of reasons anyway), and many labs pre pour petris then gamma sterilize later. Also his glass petri isn’t cracked, his agar is.
