You can use a piece of stem or cap to start a tissure culture, but you must have a good sterile technique. If you only have one cake, I would reccomend that you allow your caps to reach the point of opening up "flat" and take prints on all of them. Afterwards, just dry them and consume them as usuall. You will have 12-15 prints (just a guess) AND fruits from this batch. From this point on, pick all of your mushies before the veil breaks, except maybe 5-10 per batch (when casing in 4x8 tray it is easy to get 50-70 shrooms per flush) for printing and then you are on your way.

If you did want to start a culture, it is better to use agar plates. In a liquid medium, you cannot propagate prime growth patterns or isolate contams.

no, the stem doesn not contain spores. you can inoculate a single cake, wait for the mushrooms to grow, allow at least one mushroom to break its veil, and then use one of many techniques to extract the spores from the cap and make a spore syringe. i believe one of the final topics in the mmgg discusses how to do this.

You can use BOTH techniques if you wish.

Technique #1 Would be to wait for the mushrooms to grow and then use a TEK (found in the Grow/Find Section) and take spore prints and then make syringes.

Technique #2 is to pre-prepare some substrate to use, then when the mushrooms are mature pick a nice healthy looking one (do all of this under sterile conditions) and cut the stem open. If you wish you can blenderize the piece (in a sterile blender) with some water and use the resulting mycelium water to innoculate your prepared substrate filled jars, or you can just take the section cut it up and place it into your prepared jars.

Mushrooms are merely densly packed Mycelium (for the most part). If you use steril conditions you should be able to use both techniques. Cut the cap off the mushroom and take prints and at the same time, take the stem, find a nice clean healthy section and use that to innoculate more jars.
