Does mycelium only colonize the top of an agar dish

Am I supposed to wait until an entire dish of agar is colonized to transfer it to grains, or only once the top is covered?

Before the mycelium reaches the edge of the dish.

What happens if you transfer myc that has been on a plate long enough to grow a mushrooms?

Got some plates of clones I never got around to transferring...

Can you potentially use them to Inoc grain, or will that increase chance of Contam / bacteria ... ?

I have 3 plates of tissue sample that I should have transferred to fresh agar weeks ago and never did. I'm thinking about putting them to grain and spawning them and taking new clones from those and then transferring to agar.

It is colonizing into the agar, thus why food colouring appears to vanish as more surface area gets colonized. To me that alone is enough for me to believe it is colonizing into the agar. To top it off, we have hot pour teks meant to clean myc up because myc will colonize through the agar, while bacteria will have to go around, freeing the myc from bacteria.

Why not make a transfer from those plates? If you noc grain and the myc on agar is bacterial you're giving the bacteria a fighting chance. If you plan on cleaning up tissue on agar it'd be no different to clean the plates you already have, except it'd be now and not next month. Just a thought. The plate looks clean, just dont transfer an edge since they're touching the sides. Best of luck

Mycelium absolutely does colonize fully down into the agar.

Mycelium isn't white, it's 'clear'. You see it only because of the way it messes with light, like snow and polar bear's fur.
ANY mycelium you see is "aerial", outside of its substrate, looking for food (or fruiting sites) beyond where it's currently growing.

Mycelium always grows outward in all directions like an orb, and that orb growth will appear anywhere they can grow. It's exactly like if you left a single bit of mycelium undisturbed in a nice liquid culture, except it's a fixed gel. A shallow pan of agar like a petri dish makes their growth like a disc section sliced out of an orb.  If you ever make something like master culture slants you may be able to more easily observe how mycelium grows in that orb shape down into the agar, and down into the wood stick, away from the surface, safe in the fibers of the wood, protected by the surrounding agar.

Over-colonized plates sometimes have a bit of mold or other contaminates hiding in them. Sometimes over-colonized plate look great but have lost their vigor.

Still they usually do work fine. If it were me, I'd take a couple agar transfers and put the balance into a few grain jars.

Hey thanks man.

I'm so lazy.

I just prepped some jars though so I'm taking one of these probably month old colonized agar plates and putting it to grain tomorrow.

My next move will be to make some agar and transfer the remaining month old tissue samples to clean agar.

I actually have a bunch of clean transfers in my fridge, but I haven't used this clone yet, and I only have two plates, one in the fridge, one that's been sitting here for like a month. I'm going to use the one that's been sitting here for a month.

I just spawned 7 jars I inoculated with one of his brothers who was extremely old as well, ill see how they produce.

You can take sections of the stem butt and get fruiting genes right away, before the agar pin sporulates. If those plates sector, take the strongest growth. Some of my plates grow down the agar and up the glass after several months in the refrigerator. It's not an ideal situation.
