inoculation on wild spore print
    #24842749 - 12/12/17 07:52 PM (7 years, 1 month ago)

I recently inoculated a petri dish using an inoculation loop from a wild spore print. There is thick growth where I dragged but there is also minor growth around the edge's as if my flowhood blew some spore's around. Has anyone else here expirienced this?

I have . Whenever you tackle a wild print you must know that there will be some sort of tag along spore to cause problems for you. It is just another transfer for you to make .

whether or not it's wild has nothing to do with satellite colonies on the edge. which you shouldn't be getting with a FH going from print to agar.

You don't think that the FH could have blown spores off of my loop?

I have not had it happen. I would say it is POSSIBLE though. Do you cool your inoculation loop in the agar which makes it moist and holds the spores better?

Yes I do dip it after sterilizing. I use little mason jars instead of petri dishes maybe the extra tall walls have something to do with it.

Satellites happen in front of a flowhood.  There is no rule saying that they don't.

Important thing is get a few transfers from the main germination ASAP, just incase those satellites turn out to be mold.

I actually did and the new dishes have some satellites as well, I took a few wedges and put them too cardboard and those are are doing really well. I wanted the mycelium to sector but so far it has not its kind of just growing out into a velvety circle. It is Azurescens. Is it normal for it to not have any rhizomorphs?

When that happened to me it turned out I was transferring a mold that doesn't make colored spores.

It's taken to the cardboard, how can I find out if it's really mycelium. I've never tried a wild spore print only cloned wild specimens. I've read that Azurescens doesn't rhizo.

Well did it start out really close to round. I ask as mushroom mycelium will start out od shape due to the different sectors growing at different rates, while mold tends to start out round and uniform. This isn't a 100% accurate way, but can give some supporting evidence.

Hopefully you transferred mycelium and the mold was left behind.

I transfered as close to the inoculation point as I could went for the thick white stuff. The Azurescens mycelium is very dense but also has some Arial tufts it looks like pure white Rabbit fur and I kind of thought maybe some of the more subtle mycelium strands became airborne from the air flow of my FH and blew elsewhere into my dish. Sound possible?

Yes.  When transferring away from contamination you want to use still air methods.

If I am going to make cardboard spawn then put it to wood chips for an outdoor bed, is there really any point to me cleaning up the mycelium on agar? I originally wanted to take a few sectors isolate them then spawn them to their own separate pots too see which one fruits the best next fall. But the outer edge seems to be pretty uniform. From the pictures I've seen online in my opinion the azurescens mycelium looks to first go out tomentose so the rhizomorphs have a nice bed to crawl over?
