Effect of gravity on agar plate?
    #2482538 - 03/30/04 01:07 AM (20 years, 1 month ago)

I have very recently started experimenting with agar. I have a second generation agar plate, that I have kept inverted since mycelium transfer (3 days ago) so as to keep condensation to an absolute minimum. Though the mycelium looks very healthy, it seems as though the growth is on the surface of the agar only, and does not penetrate it. I am just wondering if this is normal and OK, or if gravity is having an adverse effect in that the mycelium does not want to grow up "through" the agar.

Any insight or help with this particular topic would be greatly appreciated.

It is perfectly normal for the mycelium to remain on the surface of the agar and it makes no difference if the plate is inverted or right side up. Inverting does help to keep condensation from dripping on the agar surface.

Excellent, thank you.

I had assumed so, but from some of the pictures I have seen of very healthy looking mycelial growth, it has always appeared to be growing underneath the surface of the agar.

Maybe when the photos are shot the dishes are held tot he light, so the culture looks as if it is below the agar
