They do.
Letting them sit a month is a good test of if they're done right. They should be drying out before contamination

Idk why everyone is so fetish about layers of micropore tape tho. If the tape is good enough to work one layer would work. Unless it's for durability. Stacking filters doesn't make them more effective

thanks guys..its weird because they're all in un-modded half pint jars, must've botched the cook by letting the pressure drop for a bit midway or something

Been thinking it might finally be time to use disposable plastic petris, they look so nice and clear.

if you shop around a box of 500 60mm dishes is about 50bux. the 60mm are about the same size as a miniround. 400ml of agar can pour about 60-70 dishes:thumbup:

I'm fetish about lotta layers and tiny holes :hillbilly: 

You can keep them like forever in a tote that way, not even the condensation will go away after a month if the containers are smallish like the ones I use.

Just one layer of mp tape sketches me out, a simple scratch could ruin the filter. :archiebunker:

How many petri dishes do you throw out on average per month? Just a rough estimate?

I pour a sleeve of plates every 7-10 days. Should average about 1k plates a year.

Looks like I might have to do some pH testing with cubensis to establish what type(s) of cellulase it has. I found today there are 4 types.

pH 7 seems to be ok, however I am inclined to think its a hybrid cellulase, as dung is usually acidic.
Here are today's surface shots, out spread was better on 1.2% nitrogen (no starch-sugar).

The cubensis is officially colonizing 100% cellulose areas, and pushing better.
Not an ideal scenario for trying to make a cake, best used with spawn.

However, germination and transfer to agar is possible.

   

The birch wood peg has a pH of 6.2.

I'm wondering if I'm growing a jar of mycogone. This jar is really confounding me. I'll have to post pics when I get back from the weekend or later today if I get time. Whatever it is it's evil. No multispore jar starts on the 11th and is halfway colonized. I have seen a lot of contams but this one takes the cake for cube myc imposter of the year.

I've updated my pocket guide with enzymes and pH, and re-arranged some text for better reading, should take only 20 minutes to read.

I also updated my T-Gel agar recipe, and added images-data on the end pH, and alternatives.

Tea + water, can attract mold, it's not 100% proof, mold is lignicolous (wood loving).
To be used as the source of carbon + nutrients, bonus inhibitory effects.

Non-lignicolous mycelium will require a carbon source.

I suggest the 1.2% nitrogen recipe for use with the cellulose agar alternative.
You may need to adjust the end pH to suit your mycelium, enzymes.

Unless you are using starch rich grain, and pH 7-7.5.

Cubensis var. Burma, dry spores onto a peg, and a spent tea bag.
Running on just cellulose, totally inhibited, pH 5.8-6.0.

Right: Lignicolous cyan sample, also on tea.

What is the best way to acidify a pH 7 substrate, that I can get easily and cheap?
Could vinegar work, or will it do something weird to cubensis mycelium?

I would like to test the pH level for cellulase, by cubensis.
I've seen people put cubensis on card (acidic).
