I am having a hard time duplicating this...

1/2 cup water 2.5 grams of lab grade powdered agar from a reputable vendor and 3.3 grams of powdered potato flakes...

It just gets thick really fast and when I pour it, I loose so much on the side of my pot I can't hit all 8 mini rounds. Then it never fully hardens...its stays about the consistency of thick mucus. It will smudge all over my finger like snot even after fully cooling.



I even followed this video exactly and can't get it. Mine comes out nowhere close. What the hell is going on? In the video, his is still the consistency of water and appears to pour easy.



In the video the stuff looks like water and pours in easy. Mine is thick and viscous, but never solidifies. I am very confused.

is your agar specifically just agar powder? or is it perhaps a nutritious agar premix like mea or pdya?

what kind of potato flakes are you using? plain or flavored?

try adding more water. dont forget to add a sugar source.

It is pure lab grade agar-agar...the potato flakes are not flavored. I got the ones that were basically dehydrated potato's with a preservative or two iirc. I did add a drop of kero and some food coloring.

I am super baffled...

Not yet...i tried less agar though. I did like 2.1gr agar to 3gr potato and 1/2 cup water. Waiting to see what happens. If it does not work I will try again with the original recipe and more water. How much more do you recon I may need?

Did you heat it up all the way till its nearly boiling? You have to make sure the agar powder has been fully dissolved and melted.

If it still doesn't work then your agar powder is probably just shit. Go to a supermarket of the asian variety, and buy the packets of agar powder there. A good one is the Telephone brand one. They're about $2.50 a pack.

The agar is a few years old and was sitting outside in the garage in a plastic container with a screw on top. It looks and smells good/dry. I was wondering about that though...

Is your water really acidic? A low ph can sometimes prevent the agar from setting properly.

Here are the great value/walmart generic screw top mini-rounds I got for this. They work great and they are very small - 4oz / 1/2 cup

About 2.70 for a pack of 6 - located in the aluminum foil isle.

Since no lid modification is required they are a breeze to prepare and cook up

I have used several sources of water...I ordered some new agar just in case. Going to try the telephone brand. I will report back.

Those are gonna kick ass you'll see. I'd personally drill a tiny winy little hole through the side and cover that with 3 layers of mp tape, so you don't need to foil them during the pc cycle and the tape will come out next to dry when the cycle is finished. You can also reuse the tape for the next cycle, just simply add an extra layer of tape.

Personally, I would find it to be more of a hassle having to unscrew the lids a little before the pc cycle and then tighten them when the cycle is finished, especially if you do lots of agar. The tape takes care of that. Maybe it's more of a hassle at first (drilling holes and taping) but not in subsequent cycles when you only need to add 1 layer or none at all when there are enough layers from previous cycles.

I did my first 6 already about a week ago and bought another pack to have on hand. Not doing a ton of them at this point since still seeing how my first attempts turn out. I didn't put foil on them, just made sure they were sitting outside of the water in PC and stacked on each other. Waited until they were all cooled and just tightened the lids as I took them out. Since I have to pick them all up anyways, its literally no extra work to give the lids a slight tighten at this point.

I put clone tissue samples to 3 and a drop of spore solution to the other 3. So far the spore solution ones are showing a small spot of growth where the drops were and looking good.

The tissue sample ones are getting white fuzzy on the tissues, but one of them had 2 small tissue samples and the 2nd one looks to be showing some contam unless its bruising. Its a bluish color and in this particular dish the agar came out kinda lopsided and its bunched up to the side of the dish, I guess because it must have became crooked during PC. The contam looking spot is in the side where there is basically no agar so I was wondering if this would cause the growth to bruise maybe? Guess I should take a pic... not sure if bruising would even be something you'd see at this stage of myc growth though.

Tissue samples will fuzz over and turn blue.  As long as it is where you put the transfer, wait and watch for growth.
