I put a hole in the side of those big lots containers. I've also kept some cultures going for months though for stones or pins

Good to know Saturn Iβll keep that in mind. If it helps at all though Iβd like to use it.

For clarification, are you saying you didn't use any karo or honey? Or did you still use honey? Cus its an either/or ingredient rather than both

He probably means that he omitted any sugars, and only put agar and potatoe.
MadHatter, most people use Karo light corn syrup, I think. But I've only ever used Walmart Great Value brand light corn syrup (lol) for my PDA agar. Honey should work fine as well, though some people avoid it because of its supposed antifungal properties.
Between the two, I would choose Karo, but if you already have honey and don't have Karo, then you may as well give it a try. It'll probably be fine.

I think Iβll go with the Karo if itβs what most people use. Itβs not to expensive anyway and Iβm sure I can find some other uses for it. Guess itβs not that important but if it gives me a edge why not? Thanks again guys. Look forward to starting my agar adventure!

IME the opposite is true. Less nutrition has lead to zhizomorphic growth.
making it much easier to distinguish from foreign growth on plates.
Try using it both ways so you can see for yourself.

Hello Pastywhyte, I brought some small mini rounds similar to the ones in your pictures, how do you keep them from melting in the pressure cooker? and is the hole that you made in the top use as a air hole and inoculation hole? Thank You

If your containers are pp5 they won't melt... And that hole is to allow for air to escape the container during pressure cooking, with no hole the containers warp or they get crushed by the built up pressure inside the pressure cooker

If you're thinking agar plates have an inoculation hole then I highly recommend more reading.. And also the fact that he doesn't mention anything specific about keeping them from melting should be a clue.

My first Pasty-plates are cooling off in the PC right now and I'm planning on trying my hand at cloning with them. This will be my first attempt at both cloning and agar.
If I've understood correctly, the best way to go about cloning is to select a good fruit, flame sterilize a scalpel and then cut some tissue from inside the stem in order to get a tissue sample that is a bit more sterile than the outside?
I've also seen it said to use the portion at the base of the stem or just under the cap.
I won't be using a pin as the fruits I will be selecting from are past pinning.

you want to pull the mushroom apart with your fingers then take your sterilized blade and cut a small tissue sample from the stipe and transfer that to agar

I appreciate all the help and information that you have posted. I have some questions about THIS LINK >>>> https://www.shroomery.org/forums/showflat.php/Number/19208976/fpart/1/vc/1 . My question is when and how are those plastic mini rounds in the pictures are inoculated with the spore syringe or shroom samples. I see when the holes are drilled in the mini rounds , but I dont understand how or when the spore samples are inoculated into the agar, can you explain that step. Thank You.
