no. i chop the base off to about an inch stump. then i pull the base apart like a bag of chips.. if that makes sense..
exposing making 2 halves. i then take the flamed blade and gently scrap out a sample.. put to agar. ... eat the discarded specimen

So this shouldn't be done in front of a flowhood because moving air is bound to blow whatever is on the outside of the stipe into the center while tearing it open? Also doesn't tearing cause cells to break free from the outside flesh and be flung into the air anyway?

even though ive had clean clones on the first dish, id be way to scared to shot tissue into LC.
put to agar and then poke with the needle would be the way id go. ...wait. i did go that route!
scraped out tissue from the guy on the left.
agar'ed
poked, LCed, poured, colonized, spawned
i should of cleaned up the original culture. but fuck it. it worked too easily to care. and the tiny specs of myc from the poke slimmed down the genetics even more so maybe you dont need to use a super nice culture.

The slimming down of the genetics is a huge one I tell ya, it trumps the 'easy' aspect of noccing with the poke.

What about poking a pin on agar?

Depends... If you've taken transfers from that plate, I would not risk it. If you work with snap-on pp5's, I wouldn't. If you use half pint jars or petris, maybe. But I'd personally shoot the pin tissue on agar first, no matter what. We're talking days till you can use that plate to inoculate.

Rip in half with your fingers and then use a flamed scalpel to take a nice clean sample from the interior of the stipe. If you cross cut, it will drag all the filth on the outside of the stipe into your sample area. At least that's how I did it....Quote:

Also forgot to mention Van, sometimes it's just easier to transfer the whole pin to agar, them skinny pins are not that easy to poke sometimes. Then let the pin colonize the plate a bit and take a biopsy from the myc that grew from the base of the pin, as it is always more vigorous.

Right. Well i saw eat colonize a 1/2 pint brf cake in 3-4 days using blended stipe tissue. Is that faster or about the same as using LC/LI to inoculate a 1/2 pint brf cake?
