In four days both jars (turkey tail and B+), are loaded with beautiful cultures with the Josex Poke method....
This is a great way to eliminate contaminates in your Lc's
I had some doubts at first.... But not now....!

4 days I used the method with some brf plates (josex idea) nice white growth coming in.

made some ape broth with the poke
using the blunt tipped needle is more of 'core' sample taking. both ways work. i just happen to have spare blunt tips

Do you mean pf tek pucks? That's gonna get you a nice clean culture but if you want bad ass growth try cvg+brf pucks, good to test clones/cultures or snatch some clones when they grow out, it also has some 'cleaning effect' if a culture does not look that pretty for whatever reason.Quote:

Noice! Dude I wish I had petris just to take those kinda pics, have some biopsy action on agar right now, trying to get some isolates out of some clones (useless I know) just for the fuck of it. That's some wide ass needle you got there. I find it strange you can actually take a 'plug', coz I tried with a 14g blunt needle and could not see anything in it. When you poke some myc fragments will rub off on the needle but it can't be seen. I compare it with putting your finger on a glass, you aren't going to see anything but lots of skin cells will be there nonetheless.

Curious, when I started doing this I slso thought the needle had to be kinda warm for it to work, enough without killing the myc, but a cold needle will work the same.

I'll throw some cvg in next time as well.
One culture I'm trying to clean up the second one is for fun.
Going to try this with lc when these plates grow out (if they dont comtam).

I got 5 jars, 400ml each of some diluted grain water with a pinch of dextrose for the fuck of it. Got some clean LGT clone plates as well. Tomorrow I will inoculate some LGT and Blue Oyster LC jars.

Never use pucks to inoculate your LC, let alone the one you're using to clean up your stuff. You won't be able to inspect the culture easily with these and they can hide nasties. If you think the pucks are clean go ahead and poke them to transfer to agar.

Different plates would be used for the lc. Should of specified.

Can inside stipe tissue harbor bacteria? If not, I can see how using a flame sterilized blade to cut the butt off the stipe and using a flamed sterilized needle to collect inside tissue to noc an LC directly without going to agar first. I know it is not common practice but it could work well if the inner tissue isn't compromised in anyway.

I don't know, maybe. I always shoot the tissue on agar first just to make sure. It's a tradeoff between waiting some days to test it on agar and shaving off some days but without any guarantee the LC will be clean.

Cutting the stipe with a flame sterilized blade is still bad practice. Even thought the blade is sterile it can still push bacteria from the surface of the fruit to the inside. You will have much higher success ripping the stipe

How do you rip the stipe? I'm a little confused? You do not use a blade to pierce into the stipe?
