So, I impulse purchased this microscope and these vital stains today...

I'm not totally sure why, but it seemed like a good idea.

My Plan
Figure out a way to compare vitality between clones and isolates without needing to spawn them out.

Thoughts?

Vitality testing is an interesting conundrum. How do you do it without killing the cells?

For brewing yeast viability determination. You can do microscopy with methylene blue. Dead cells are assumed to absorb it so blue cells are dead. You can get a percentage this way.

More accurate ways use probes with sensors that measure radio frequency capacitance and fluid conductivity. The name brand in these os ABER. Whos products cost as much as a new car.

Took me a while to find it, but I assume you're talking about the CountStar and PerfectPitch? Whoa, never heard of these but I already know any company that doesn't list prices on it's products will not be covered by my Prime membership.
And I'm more of a Computer Scientist than biological scientist, so this will be an interesting project indeed. Probably easier for me to write a program to count cells from a jpeg than correctly stain a slide.

HOWEVER from my limited research into the matter, methylene blue only stains dead cells and is presumed to be a vital stain for the most part. I've also read how it acts as a nucleic stain in some cases, but until I start my own trials, I don't have much to say on the issue.

I posted this here mostly looking for tips or suggestions of what differences you look for in your own cultures... rhizomorphic growth, even distribution, defined veins, etc? This will probably help me devise a starting point for my own testing.

I keep thinking about the ever growing cordyceps market. :philososloth: Cordyceps on the brain. Cordyceps videos. I will do this and it will be great! I also know where to purchase nice cordyceps cultures too when I have the money.

Does anyone think I can give bugs cordyceps. How would this even work. I must figure it all out. I know you can grow it with the right combination of supplements without the bugs. But wouldn't it be cool if I could grow it on the bugs and just watch them.

Ooh- Dialectric Microscopy.... That IS intriguing.

More radio frequency stuff that maybe might involve mushroom cult. Definitely more high-tech than mini-lightning zapping one's mycelial mass.

Awesome.

P. Stamets would have a cordyceps-based bug-killer on the market already if wasn't for those pesky competing gazillionaire pesticide mongers and their influence over the industry, IMO.

Also I just remembered that bugs are creep and zombie bugs are even creepier. It's such a peculiar notion that fungus can get the insects to do their business.

I found this video which is great but I have no idea WTF they are saying. Looks like some kind of nutrient broth over sorghum. It might be Thailand. If anyone knows what they are doing please tell me. I am willing to trade nudes for this information.:grin:

One of the supplements used is derived from silkworms. Some kind of silkworm extract. Unfortunately I can't understand the videos. I'm sure you could. I need to know what the magic broth is.

They can be grown on whole brown rice enriched with certain nutes. I have militaris and cs4. I did a cs4 grow on
whole brown rice using said enrichments. When it was fully colonized it smelled very strongly of ammonia and a quick
search showed that the cs4 culture produces metabolites that smell like ammonia quite frequently. I didn't actually
use any of it cause I was still a little freaked out.

Militaris can be fruited on brown rice with certain vitamins added. No bugs needed. My agar recipe for cordyceps did
include ground up crickets though :lol:

Using my microwave pressure cooker grain prep, I decided to skip the hob pressure cooker.
I'm testing to see how long it takes before I get any visible contamination.

So far it's been 5 days, and there is no visible contamination.
I'm using organic rye that has not been treated.

I added dry spores for a timescale.

We all started noob!

I will be happy with 7-8 days, seems the first step works well for a no soak gain prep at least.
In comparison to a hob boil, I maintain more nutrients, some pressure and steam.

The microwaves themselves are just a bonus, and not calculated.

You might prefer a standard PC used in the same way.

There is also "easy cook" which is partially cooked grains, however they have a lower cook time.
I managed to pressure cook some easy cook brown rice for 28 minutes, packet dry.


Some interesting notes:

Brown rice releases starch very easily which is amazing for spores.
Rye is much firmer, and takes much longer to contaminate.


Brown Rice:

+ Spawn > cellulose get's an amazing reaction.
- Prone to many types of contamination.

Organic Rye:

+ Does not leak starch, it's firm.
- Growth is slow to start.


Rye has a higher spread (colonize) reaction, rice has a slower well fed reaction.

This is all theory based on observation.

I don't know how you can measure nutrient retention without performing calorimetry. How do you know something retains more nutrition?

Says what? If you prepare with different heating methods theres different nutrition?

Theres always the company scientists who will say if you cook food in our device more minerals and vitamins stay in your food. If thats what you mean then :shrug: lol

No but it is standard knowledge bod that boiling foods depletes its nutrients. For example, boiled vegetables are
useless for nutrients
