What the hell ill give it a try too next time around. I like it. Though big wedges to my LC jar with some glass in the bottom and they're ready to use 3+4 days later with one agitation per day. I still like the idea behind this. Nice one josex

This tek is not about taking a perfectly visible core sample either but about inoculating with literally microscopic pieces of tissue from a clean agar culture to make the procedure as safe as possible, so you can perfectly get away with no testing and dicking around.

Also, I would not do this method poking an invitro agar pin as Cron said, I'd transfer the pin to a blank first. My confidence in pp5 containers with snap-on lids has shaken quite a bit after my last mold disaster a few months back. They're far from perfect and shit can def get in them if you keep them for long till they pin.

So, I had a transfer that landed with a 45° angle on top face. With nice ropey strands. 5 days in it's radial but the top launched up with 2 twisted pair looking strands about 4cm long into the air.

Decided to use flamed tweezers and try an LC with the strands. Looking decent so far. Inspired by this topic. Thx :cheers:

Because you can't see through the bottle. Even if it was clear plastic, visibility would be horrible.

The idea of pouring the LC into the bottle can be intimidating at first, and you don't need to do it if it makes you nervous.
I only use it when I make a 400-500ml LC to inoculate lots of jars, enough that they won't fit all in the SAB.

If you plan on doing lots of jars in one go, then yes, the bottle comes in handy. Or you could also make several smaller LC's to inoculate lots of jars in the same session without having to use a blender bottle. Whatever makes you feel more comfortable.

ive been using quart jars with my usual tyvek mp tape filter.

500ml of liquid in a quart still leaves plenty of headroom to stir up the broth w/out getting the filter wet.
and its super easy to pour. but i like the idea of the really small spout to pour from. less exposure=less chance of ickies.

FIRST LC KNOTS.

cant see any but ya know.. they dont show up with my shitty nokia fone.

yah dude. i got 3 more quarts to spawn tonight.. and that bag should be done by the weekend. mono town:borfase:

..and ive done about 3 rounds of this. lots of jars. lots of broth. no contams:thumbup:

now pimpin is easy :snoop:

As far as GE is concerned you'll be fine. I really think you can even screw the lid all the way during colonization, LC's don't need that much GE, if at all.
The part that sketches me a little is having to unscrew a bulky plastic lid to inoculate the broth, but then again I'm a paranoid fuck when it comes to noccing LC's.

We'll see how that goes :popcorn:

Hey CBK, I think I remember you posting some nice wide pp5 tubes with screw cap, right? If you still have them around you can use them to mod a lid for LC's for easy inoculation and GE. I'm not home right now, but can tell you later if you want.

I have the metal lid unmodified and upside down.  I've had issues with LC and plastic lids in the past.

I'm not actually sure what you're talking about tbh. Ziplock twistnloc?
