I'd say lc is the fastest easiest way for noccing cakes.
I wouldn't risk the stipe tissue inoculation in anything but agar.

for me using a thick lc is about 4-5 days with 15 ccs poured over the top and down the sides.
I've gotten 3 days before but the growth is light untill day 4 or 5.
Either way 3 days vs 6 days isn't going to change much. just make/inoculate the cakes ahead of time so the couple extra days isn't going to be a problem.

Yeah LC to cakes is really fast and me loves some invitro cakes from LC.
This is the one I'm most proud of:
KSSS first flush.
Same cakes on the fourth flush:
Regular looking KSSS it was always the same with this culture after the second flush.

i used tp go directly with inner stipe tissue to pf cakes all the time. that last tyc grow i had was from tissue stored in the fridge since 2014. but i blend it up into sterile water then suck into syringes. store or inject directly into pf jars. but the lc will have to grow a little first before you are able inject it. so direct stipe inoculation is faster because it begins growth by the next day compared to waiting for an lc to colonize a few days then inoculate.
here is one example of inner stem tissue blended up and stored in a syringe for a few years since 2014. then grown out about 2 months ago.
nature is pretty effin amazing. its an old method called 9er tek cloning method. i never go to grain with the stem tissue though. only pf jars. and all this was done in a shmuvbox lol. so it can be done
generally if you inject 5ccs into one pf jar it takes about 3-5 days for full colonization. if you do a few cc's per jar it may take up to 10 days like shown below here.
i grew like this for about 8 years consistently with little to no contamination. i could turnover a clone in about a 2 weeks time to fruit.

Yeah mike, do that with your LC, see what happens, I for one will be watching. That works for cakes and it's a whole different method.

It's not hostility, it's saying things as they are without beating around the bush. You seem hellbent on doing that and it's all round awful practice, instead of doing what says in the op, that has not failed me a single time yet. Half assing with LC won't get you far and I don't get why you want to do that instead of doing it the safe way. There are noobs watching this and they might think what you propose is the way to go, don't want any fuck-ups coming from bad info in this thread and I'll not hesitate to call out bullshit when I see it if that can save people who don't know better from fucking up. Nothing personal man.

Well someone brought up the idea of ripping the stipe open with their unsterile hands and I really just wanted to know how that is better sterile technique than using a flame sterilized blade. Instead of an answer, I got the old "You are derailing this thread" response.
I am not at all against your tek in any way, shape or form. In fact, I think it is a great idea.

that has nothing to do with the thread. and I think the answer to that question has been answered on the shroomery countless times. Cutting brings in contams from the outer tissues into the middle where you want to take a clean cut from.
Ripping is done with gloved hands and in the sab and carefully so you don't have your hand going over or touching the clean inside.
ok enough about that.

was poured lc. my first bag in the sab that hasnt molded. sweet.
i think i gt my bag pouring down. tips welcome on that one. the spawn was iffy. soomething might of fallen inside during inoculation. smelled and recovered fine looked like shit though imo.
spawned to a brick of coco. first mono in awhile.

Hey Josex, I'm waiting for the syringe to cool, I'm about to nocc up grain water with a clone culture. I have a small jar with SHIP and Whatman. Would you still perfer that I lift the top and squirt it in or going trough the SHIP? Everything is 120% sterile

You mean grain water like in the op? Nasty sediment and all? That could clog a syringe fast and furious I think.
Inoculating through the ship, that's a no no. Take into account that ships are usually pierced with a flamed needle, still hot, and this is not the case, the needle will be cold when you inoculate. Peeps kinda rely on the needle being hot to hopefully kill some nasties that the needle might have dragged from the ship. Also, you can't sterilize a ship, you sanitize it with alcohol.
So yes, I'd advice you to inoculate by lifting the lid.

My grain water was filtered a few times upon making it, then frozen and after defrost today I filtered it one more time. The sediments are barely visible, pretty clean I must say. The jar and lid with the SHIP and Whatman came out of the PC covered with foil and the foil was removed a sec before inoculation. I told you everything was 120% sterile
Btw I already inoculated trough the SHIP, and this LC is more to test the GW and I have no plans (as of yet) of inoculation anything with this LC. This was just to test out your Tek trough the SHIP and to test GWLC. I'll post results in a week
Edit: I poked the SHIP one time before with a flamed needle and never again. Now I just sterilize the syringe and go to work. SHIP looks brand new still.
