Currently working on reviving some cultures from fridge storage after taking a break from mush cult. I have one clean plate from a culture I’d like to safely store, and also begin expanding from. What would be your preferred workflow in this situation?
I’ve made a single transfer to a new plate which appears to be taking off nicely. Provided it remains clean, my plan would be to transfer to additional backup plates, and a couple smaller grain masters to store and expand/work from.
Hesitant to send the original/donor plate directly to grain, as a couple of my fridge plates grew visible contams over the months they were stored. But planning to hold on to it in case it’s needed, or until it gives up a plate pin to clean up/store.
WWYD?
Good instinct.
Run your t1's or t2's. A plate or two is cheap compared to a month and a half lost.

For cultures I cared about, I liked transferring to 2-3 plates if it'd been stored for a bit. I'd often have a plate with 3 small transfers on it and the others 1x each. When coming out of the fridge, I'd never go directly to anything, especially not if fridged plates showed contams.
The fridge doesn't just slow mush mycelium, but all growth. Bacteria or mold spores might be lurking, but not exploding until the right conditions. So get a few plates from the original, let them grow for a bit, and take it from there.
If everything looks good, I'd fridge at least 1 of the 2x1 transfer plates and expand the 3x one, if clean. But that's my style and demands back then.

Thank you both.
In retrospect I should really have transferred to a few plates at the time, not sure why I didn’t tbh. Made a mental note for next time.
This is ~6 days of growth, probably a bit of delay at the front end of that in bouncing back from fridge life. Gonna let it do its thing for a few more days.
After the window of time where bacteria/mold would show - Is there an advantage to letting this T1 grow out all the way before transferring it to additional plates and/or testing on grain?

I banned myself from this thread years ago because I kept accidentally answering questions...
I can do that now!
I try to go to grain before the culture hits the edge, but I'm not sure if that's paranoia or not.
as for going to another plate it doesn't matter if you do it right after removing from cold storage or if you wait. imo the only benefit to waiting is improved recovery time.
just don't take a plate from cold storage to a grain jar without testing it between.

Thanks. Yeah that was transferred from the fridge plate, let it ride for a couple days and then sent it. Seems happy

Yo Beef and Tryp, those badges looks good on ya.
Just got my shit set back up and acquired a bunch of swabs. Looking forward to spicing things up with some new varieties as I’ve been running the same few cubes for a minute now.
Thinking I might make it my goal to run a bunch of stuff from spore on a shoebox scale and make a shitload of swabs for either the apocalypse or a giveaway, whichever comes first.
How would you suggest organizing my agar workflow/labelling etc to run multiple varieties this winter, without it becoming a chaotic clusterfuck?

Im afraid Im right there with trypt. also, same re: mental health. im already struggling with knowing what i should prioritize in terms of genetics. I try to limit my workload but one thing i have found that really helps?
send a ton of it to someone else you trust for them to help you work through the genetic material! haha.

Same tbh, I’m glad I’m not alone.
When I most recently tossed all my shit and threw in the towel I just had too much on the go and didn’t have the mental energy to care anymore lol. I inevitably get excited and bite off more than I chew in all of my pursuits in life, and then get frustrated when it creates issues for future me.
Being ambitious with hobbies feels kinda backwards but I know at least a loose goal is necessary for any degree of organization or planning to happen inside my brain. I think just running a bunch of stuff spore to spore and not losing my shit seems like a reasonable goal.

I figure you should start slow with one or two varieties, get em to grain, and cycle a new variety in right around that time.

There's a book that had a huge impact on me: "The Paradox of Choice: Why More Is Less - by Barry Schwartz". The gist of it is that the more choice/options you have the less happy you are.
I read it more than 10 years ago, this concept is now carved in my brain. Granted I still have to fight it every week, but now saying "no" to a new project/species - even if hard - feels good for my mental sanity.
My 2 cents: select 2-3 species MAX. Grow them from spores to prints/swabs - even better I'd put them in cryovials filled with distilled water = so easy to fire up a culture afterward and lasts 10+ years apparently; once it's in cryo, get rid of everything but the cryos - and only after that you start with another set of 2-3 species.

Just read a couple vial teks and it seems like a great way to reduce plates accumulating that I will never use.
More is less applies to many things in life. I’m a more guy and get off on starting things… Closing loops before starting more, not so much. Completing things before taking on more is already a goal of mine in work and life, which could definitely be applied to this hobby.
Thanks for sharing that
