Could be a bit of both. I find with fruit development it's often a combo of lots of factors.

If you sterilized the whey any bio activity in it is dead. Pasteurization by definition means that some life is left to protect the substrate.

Sounds like a mold farm waiting to happen. I don't know what you think you are going to accomplish but it sounds like you basically partially sterilized a bunch of contam prone media.

Probably okay but ditch the heater and keep an eye on things. Watch for bacteria blooms. Heaters cause more issues than they solve usually.

You ran the PC dry. Either use more water or lower the temp so the weight isn't rattling so much. Also be sure the jars are elevated off the bottom of the pot.

Of course. You will see a gamut of results. That's why we test isolates or clones and only keep the good ones. Of course ms is a crap shoot every time.

Depending on how they were dried you may be able to reanimate them with antibiotic agar. However if they were dried with high heat it would be pointless and even if not it would be a challenge. Personally it would be far easier to just buy a syringe or print.

You can get gentamiacin to make antibiotic agar or buy tetracycline from the pet store. But you cannot pressure cook tetracycline so it needs to be carefully added during the cool down before you pour your plates. It's a bit tricky.
I don't recall hearing about your country banning spores. Should be easy to order some.

No. The whole point of isolation is to remove genetics.

Its mold and you don't save plates. If you have a culture in it you transfer to a new one. If you don't have a culture in it you toss it.

I'm not sure what the issue is. It's a spore swipe plate. Take from the areas you expect are good, avoid the satellites and if your receiving plates don't have satellites they are good to use. Not sure what the concern is

mos619 said: Eh, I'm not concerned so much as curious and trying to learn...like I've only made/seen about 5 swipe plates so I'm honestly not even sure what they normally look like. My expectation would be that growth would be isolated to just the path of the loop, which it more or less is, but there's those couple outliers I've seen on basically every plate I've done so I was just wondering if that's a thing that just happens and if there's a mechanism behind it.
We're all sorta scientists, I'm just reporting my observations and trying to compare notes
Well it's possible that they are just spores that got flung off the loop. I have had satellites from perfect transfers that ended up being cube mycelium. I suspect it was a fragment that fell from the wedge during transfer. If that can happen then it's certainly possible that spores get bashed off the loop during swiping. Lencacillium spores can move up to 2 feet when hit with a droplet of water so we know they can travel.
Or maybe it's satellite colonies from sloppy transfers. If they take over the whole plate or turn color you know they are mold. If not don't worry and just transfer away.
I am definitely not a scientist. But I appreciate your sediment.

I just save some in the bucket and top. Seems the easiest option and is what's giving me good results. Course I'm strictly coir verm so I have that option. But then again not many subs are as easy to prep as coir verm. Lazy way or highway for me these days.

Could be mutation, too much nutrition, or contam. Time will tell.
