Nest scientific has 500 for $87 plus shipping I believe. They had a deal on Amazon recently and I just got 500 for $47. Couldn't let it pass up lol

Hey Germs, How's the quality on those nest sci plates? I saw that deal and was debating getting a case...I use falcons as you can tell the difference in quality...really does make a difference, but they are super pricey.

They've been good so far, poured my 4th sleeve of them today. I don't like that they're vented, I've had two out of the first 60 get tiny satellite molds on the edge without me doing anything to them. They're stackable and have great clarity so for the price I'm more than pleased. Only two of the whole case broke in transit.

Once its made and stored in the fridge do u need to loosen lid before microwaving and returning to liquid?

Oh yea right on BOD i read that part. But that was about forming the agar in the first place. My question was regarding using the agar after being stored in the fridge and returning it to liquid state since its not being boiled. I should have assumed. The fear of contams is REAL! haha

it is being boiled. you basically have to to get it back to liquid. there will still be some unmelted mass in there while the rest of it hits a boil

Just starting to learn about Agar and am curious why you first add ingredients then boil in the MW, and then PC it.. wouldn't just using the PC do the same thing?

You’ll get mixed answers on that one. From what I know it’s standard lab practice to dissolve the ingredients in boiling water but some phD on here says its not needed and is a waste of time.

On the streak plates there are a couple pics u have with dark spots is that contam? then u just want to cut off that and avoid the dark spots?
Also i see streaking is the preferred method to just squirting from a syringe when using spore to agar, would the reason be just because it is neater?

Its soot or globs of spores
Syringe right to agar and you get all growth contamination and mushroom from the same spot

ah i see so the streaking increases the solution surface area of the spores so contam and mushroom will be easier to separate
