Opening LC Lab Help!
    #24570874 - 08/22/17 02:14 AM (3 years, 8 months ago)

So I have a request for technique's and or advice on working with my new LC Lab. First let me say that I have Bachelors Degree (heavy science background) a solid foot in Sterile Technique, and my last job was in an FDA Registered Lab at a pharmaceutical company. That said, I must also up-front confess that my experiences thus far have been largely in-line with the PF Tek from days of old and not anything heroic.
I purchased the Fungi Perfecti Master LC Lab with everything on it and had several issues upon delivery that were really unfortunate, but that's another subject entirely.
So I've never worked with an Eberbach or a LFH and I now have both. I had previously done my SAB work with very good results and could use some step by step pointers on working in front of the hood... I've done some video watching, but the thought of say pouring plates or knocking up dishes in a "Breeze" (sterile or not) is kinda freaking me out right now honestly.
And would the transfer from blender to glass be better performed in my SAB?
Seriously do not want to ruin many many jars with a bad LC...
Thanks in advance for any pointers you can send my way.

Without knowing how to ride a bike is more like it.

No offence or anything Op, but why would you buy a LC lab if you've never made a LC before, or even done bulk?
This just sounds like a "more money then brains" type of scenario.

If you have a hood, it's better to use the hood then the sab I'd say.
LC is very particular, and you're probably going to ruin some stuff before you get it down. With that kinda lab or not. Failure is a part of learning.

Be prepared to lose grains dude. It's a part of it.
Start small, and do pint or half pint grain jars until you have your process down. And test every LC on agar before putting it to grain.

My LC lab cost under 30 dollars.

Don't ruin a lot of jars. Test your LC and practice. Youll never get better if you dont fail a few times so get going

I have a BS too. A decade later and I suggest most people get a 4 year head start on life and avoid college.

If you want to do science why avoid college?  Where else are you going to have free access to millions of dollars in scientific equipment and people who are paid to teach you how to use it?

Had i known better i could have got my foot in QA without a degree and could have got sent to school on someone else's dime for a BS in biology when I would have gave a shit.
I used way more cool equipment and learned way more about yeast and other microorganisms that contaminate  beverages in a real lab that had to keep a business in business.

Run your hood for an hour before use  sterilize workspace with bleach . Etc

Basic sterile technique wipe everything down with alcohol before it goes in .

Operate close to the filter , never come between the filter air flow and what ever your working with.
Keep open air exposure to a minimum even in the "clean zone"

As far as pouring plates I just pressure cook the agar in a quart jar wrap a towel around it and transfer it to the flowhood to cool.  I line all the petri dishes up close to the filter. Then  alcohol swab down the jar . My jar has a simple hole on one side of the lid that was done with a nice pic and filled with poly fill before I pressure cook. It's hot, so remember to wrap a alcohol soaked towel or rag around it when you pour.  I just remove the poly fill with my left hand quickly remove the lid to the agar plate with my right hand poor and then put the lid back on with my left hand done .
Just remember to pour from the side and don't block the filter to the agar plate
.
Ps degrees or any schooling at all are not needed in this hobby. Neither is fancy equipment for that matter .
