First time with Agar

So I took a tissue sample from one of my larger penis envy fruits and cloned it. I have 14 plates, and they're starting to form white around the tissue sample I took. Is this what it's supposed to look like?

Yes. The tissue will turn dark blue sometimes, brackish, and will fuzz up and the culture will start to grow in a 2-D circle around it.

What you are looking out for is tiny white spots growing that arent growing off the culture, those colonies can be mold.

The other thing is bacteria, and I see that much more often than mold on plates. Looks like boogers, yellowish looking slime- here is a thread about it below.

Once you see enough clean growth to take a transfer from, do it.

14 plates , are they all different clones or are they from the same fruit? If its the latter , just transfer from the best looking culture, maybe a couple since you're learning.

Also, how long can I store agar in the refrigerator for? Agar that has been pc'ed 30min @ 15psi?

I could be wrong but I'd say a year or so. I've used some after a month and it still has the same texture as the day I put it in the fridge. People put master slants on agar in tubes for years and they're still viable

This is day four. I found two of the plates have contamination, but the rest look like the one in the picture. How is this one looking?

Update on my agar. Looks like we're getting some growth. When should I start doing transfers? Sorry for the poor quality pictures, it's hard taking pictures with these mini-rounds.

Ok, the only issue I'm now is that I can't see any sectors. I looked at your cleaning up agar post, and I understand what a sector looks like. But it more or less looks like what you had pictured for a monoculture.

Depends what you are going for. If you are going for an isolate, you'll have to take enough transfers to get a monoculture. If you are just wanting more shrooms for consumption, then just enough transfers to get a clean culture. Since you know it will fruit, transferring won't really affect much except for cleaning up the plate.

Until you're sure its clean. When I started learning agar I would make 2 or 3 transfers away from germination.

Now I usually transfer once after germination and if I'm satisfied, I put it to grain
