Personally i think MS > LC is a waste but agar to LC/LI is awesome. But knocking up MS grains and G2G, i feel theres a much better chance of some workable spawn than going the liquid route.
Lc is super fast when it works tho, so once you get some clean ones, its bomb. I just only got that far once and my LCs contammed a bunch of times from MS

Hey remember the clones I took that day. bro lost 2 plates to that yellow slime bacteria shit so I wasn't even trying to save those. But these. That's the tissue plates. Making some transfers tonight.
Those were made.up on the 26th pretty sure

Inoculating LC with an agar culture that's been transferred at least three times from spores is a good route to take.

I haven't really tried making one since my agar works been better. I wanna get some whatmans before I try again.
When I finally do get around to making a LC. It's gonna be of cultures that's I wanna keep because there friggin potent, fast colonizing, and funky to grow.
Not because I'm trying to prove something to myself. Which at this point however. I probably am. Lol.

Really i guess any MS culture is "dirty" more likely than not. Just a matter of how dirty. If clean= pure, 0-contam, transferred agar myc
I guess with grains, sometimes its not dirty enough to fuck up the project. Im sure theres always some bacteria. In LC made from Syringes as well

Glad to see some life on this thread again. It's been a busy couple days for mycology work, I've pasteurized enough straw for 2 5gal buckets and spawned 3qt of blue oyster to each.
I still have to do 2 more buckets for my golden oysters. It's a major pain in the ass because my main burner went out, idk if I'll even be able to pc anything on my stove now. It's a wiring issue so it should be easy to fix if I can get to the short. If not it looks like I gotta get a propane burner and tank...yay spending more money...
I've got 9 jars of atl7 coming up on the 1 month mark. 2 jars are no good for stones, 1 contamed (mucor from what I can tell) and one hasn't colonized a small section of the bottom of the jar due to excess moisture (could be bacteria too). I had a really shitty sab sess and I'm glad I haven't lost more jars. I plan to case those 2 and fruit invitro in my gh.
As far as agar work, I have 4 golden oysters and 4 blue oysters on MEA, 1 atl7 in cold storage and 4 atl7 growing out on MEA as well.
I also have an atl7 lc I started a few days ago. I tried keeping the nutes low to increase clarity but I think I should've used at least a half gram more of malt extract. I might try grain water or brf lc next to see if I like it more.
**EDIT** I just "fixed" the stove, idk why I didn't think of this before. I just lifted the top and switched the front and back element connections and remarked which was which. Works better now than it did in the first place lol

I figured I'd share this. My first ever plates I poured the other day. I have a 1000ml. Media bottle. Made PDA and I managed to pour 39 plates. Lol. Not too sure if that's good or not. Probably average. Not sure. Only had 4 plates with condensation. Couple plates had some air bubbles but not many. So all things considered. Plus being the first time I ever poured agar. I didn't do horrible I don't think.

39 plates is pretty good if you read bod's agar tek he pours 20 plates with a 500ml bottle. I rarely get 20 plates, but I like to pour a little thick cause sometimes I don't use all my prepared plates at once and some sit for a minute.

I poured ~53 from 1L last night, but usually about 50, I pour thin and carefully tilt the stack of dishes to spread the agar so they have nice even coverage

I actually only put 400ml into the 500ml bottle. 20ml a plate give or take 400ml for 20
I used to pour some very expensive media. Cost per plate at over 3$
Wild yeast media. It inhibits saccharomyces cerevisiae growth but lets other saccharomyces species grow as well as a few other yeasts like candida species. I would pour them super thin. Like get 40+ out of 400ml.

I'd probably get more plates done if my house was even remotely level. If you sit in one of my kitchen chairs and lift your feet you will just keep spinning.
How's using a 1L in a sab? Seems like it would be a little harder to handle

I use two 500mls, but I just upgraded to a FH soI recently ordered a 1L off ebay, takes up less rack space is my thought. I was pouring 50 in the SAB with only minor issues from the cramped workspace, failure rate of 1-3 everytime which I find acceptable as long as you make 2-3 transfers for every one you need..

I normally only pour 10 plates at a time. Everything I have right now is clean and I'm not working on isolates so I don't have much need for agar work until my atl7 jars are done and my oyster buckets fruit.
After I get some good oyster cultures started from clones I'm ditching the ones I have, they're pretty weak.
