More questions about Agar and starting oat jars

I have had some success with BRF (bought a kit, won't make that mistake again)... and I want to do a proper mono grow.

I've been reading/researching to death and still not clear on the easiest/best sequence to get from the spore syringes I have to inoculated jars.

Choices:
- Direct from syringe to jar
- Make agar dishes (pasty), inoculate them from syringe and then dump into jars
- do proper petri dishes and isolate?

Is #1 a bad idea? I just think that #2 involves so many more steps and opportunities for contamination..

And yes I read the flow chart above (thanks Bod!) but just not clear on the best way to achieve the "agar to grain" process.

Planning to PC a bunch of agar in the next couple of weeks and get started...

Thanks! Diving right into this now. Tyvek filter lids, silicone injection ports, cooked oats and foil.

I would never spawn MS to agar to grain, do a few transfers after going syringe to agar just to make sure it's 100% clean. it'll save you time and headache later.

Also, I highly recommend bodhi's oat tek and unmodified mono, the dude knows what he's doing.

I'm going to do some MS to grain, and some to agar, and see how it turns out.

well, I can't say I've never done MS to grain before..

just make sure it's a reliable vendor and don't use a lot of spore solution, I would only use 1-2 drops

say for example in a spore syringe of 1,000,000 spores there's 5 contaminant causing bacteriums/whatever, the most syringe solution you use the more likely it is to inject something bad into your jars. at least that's always how I understood it...

PS. TSD is a quality supplier in my opinion, If I were to go from syringe to grain it would be either a homemade or from TSD

Well there goes that idea. 9 qts of cooked oats  go into pressure cooker, and it turns out my 2nd hand cooker won't build pressure. I should have tested it before. So throwing them away and starting again...

Round 2 looking good! PC'ed the oats, multispore injected. Already shook one jar (been almost 2 weeks)

Of this batch only one jar finished and I G2G with that one to 10 more. The first 4 of that batch finished so I spawned to my very first mono. Super excited!

I do have a question about the other 6 jars - my temptation is to g2g another round, but without knowing 100% if this is a good fruiting batch it seems a bit risky. I also don't know 100% if these jars are totally clean - they smelled good, like wet mushrooms.

Anyway, should I g2g or spawn them?

I don't think going from spore to agar to liquid is bad if you have the time and setup. Then you can make your own live culture syringes and those colonize super fast. I just use 5ml honey to 100ml water for liquid inoculation solution. I cook my sorghum grain in the PC eliminating the whole precook step. something like 14oz grain with 200ml water(in a quart jar at 150psi for 1.5hr) will be enough water to steam the grain without leaving any excess water. I'm assuming this will work just the same with rye and other grains, but you will have to mess with the water level to ensure that the larger grains absorb enough water. Also you get to pick a nice thready strain if you grow out your spores on agar.

i assume u meant 15psi, i dont know if id skip any steps in the sterilization process. the soak and simmer is essential to ideal growing conditions. the grain should be dry to the touch when it goes in the jars. the soak allows endospores to germinate making them more susceptible to heat and pressure and the simmer allows the grain to retain the moisture. the pc is simply for sterilization of the spawn 15psi 120 minutes. the idea is that steam needs to hold 250 degrees to reach 15 psi. 212 is the sure fire kill temp.
