I must have really fucked up my last big round of agar transfers. I lost a bunch of plates including a clone . I don't know what the fuck happened. I'm going to get some bleach and a spray bottle and scrub that room. I cant have this shit.

If you started with spores germination could take anywhere from 3-21 days. Had some spores take even longer then that to germinate. Mileage will vary, but the average time it takes spores to germinate on agar is about 5-10 days.

This is good information. Thanks. Now if someone could tell me when this bum shoulder is going to feel better. I'm so glad they squeezed me in at pain management tomorrow. I don't complain enough so I never get any of the good rugs just celebrex which barely works.

I confess...my friend let me use their laboratory with a built in flow hood and all this official equipment, there was no scalpel or inoculation loop handy so i ended up using a kitchen knife to scrape spores onto agar plates... @mad @psilo i put those spores onto plates today under a flowhood...hoping for the best

i just made this 500ml water 9 grams agar and .5 gram of malt hoping to get some rhizo going on the ms ... im gonna have to work with the agar that powder is pretty stiff for 9 grams in 500ml

I buy my own supplies you can can light malt extract powder off amazon for like 30$ a lb if i remember and buy agar at an asian store or off ebay and yes you just add water and food coloring and heat to melt the agar and then pour your no pour or pc in a wine bottle and pour

Damn, ive been doing this wrong...i have been using pastys no pour recipe...agar powder with potato flakes. When you use the pour method how do you keep the agar from hardening in the bottle?

oh damn so your doing it right after the pressure cooker...my agar starts setting within minutes but i suppose if its high volume in a bottle it will take a little longer?

I usually wait about 20 minutes after I take it out and then pour to help cut down on condensation. You've probably got a high % of agar in your mix...2% is standard...Also, what is the agar powder you're using? I've noticed telephone brand sets up like a rock so I've backed it down to 1.5% and it's still very hard.

hahah 30$ to 3$...thanks for the correction that would have confused me whilst searching to buy some. Yeah I use telephone brand agar and do about 2 teaspoons of agar with 2 teaspoons of potato flake and a half cup of water with a drop of karo. is that totally wack?

Kinda wack...weigh your ingredients. Standard recipe calls for 2% agar and 2% nutrients....this would probably explain your issue...too much agar.
200ml of media:
Add 4g agar 4g malt extract; dog food; potato flakes; etc.. Fill your vessel to 200ml with H20 and sterilize at 15psi for 20-25min Let cool for 20 or so and pour. Letting it cool a bit will reduce the condensation build up on the lids of the plates. Also, pour them in a stack starting at the bottom to help reduce condensation as well, it's also better technique to do it this way.
