Mad bacteria and different contams can actually intertwine itself with the myc and thats why sometimes you never can clean it up no mattee what you do ... i have other cultures that are more rhizo but a few plates are giving me this

That plate is maybe a little over a week ive been transferring about the size of a nickel hoping to get away from anything that i dont want

that's a big transfer size... I use half a grain of rice... I also don't use a scalpel, I use my steel inoculation loop so that helps keep transfers small

I think I'm having a case of the oily bacteria as well, and it's not just one plate :s
Think the bacteria will hitch a ride up an agar cube with the mycelium?
I'm gonna do the agar cube in a few, but I'm figuring out what would be the most effective 1 session clean up for this. As of now, I've come up with this:
Thin hot pour over the entire media surface, followed by a large surface sandwich or warm pour with tetracycline agar. Then when it pokes through, pick up first growth with sterilized tweezers... Anyone wanna have a guess at how good this will work? Tetracycline doesn't kill bacteria btw, it just keeps them from growing, or makes them grow a lot slower.
I'm also thinking of transferring a small bit of mycelium, a piece without visible oily stuff, to a nuteless agar plate. Just demineralized water and agar. Figuring if the mycelium has no nutes, it'll colonize the plain agar plate pretty fast trying to find food, though the bacteria won't find it very interesting.. Any input?

Haha i mean when the myc gets to the size of a nickel i use a hobby knife i try to get something about a grain or smaller if i can...
I think that may work lizard ive been going up and down with my agar trying to get some different results

Which one, the nuteless agar, the hot pour and tetracycline follow-up pour/sandwich, or putting an agar cube near growing mycelium?
Come to think of it, I might even try putting a wedge of tetracycline agar next to growing mycelium, and then put a cube of regular agar right on top of that..
I was thinking AA+ too.

The whole plan a warm pour is good for killing off some nasties the tetracycline would help stop anything else the very very low nutes would help speed up the myc im not sure if no nutes would be so good but you could do like 4 or 5 grams or whatever you use for nutes

I know it wouldn't exactly be good, it's more that I want to really starve the bacteria, see if I can have the mycelium outrun it that way. Either way, tetracycline isn't that good for culture health either... I could also try a nuteless tetracycline or nearly nuteless tetracycline run. The mycelium is still all very young, it's not far from spores, so maybe I have a shot at nipping it in the but...

Do a warm pour and see how it does after that then transfer that to your nuteless agar and see how it does

Well, I'm going for a tetracycline wedge with a regular agar cube on top of it right now, I've got everything ready for that, and after that, I'm off to bed. We'll see how it goes. After that, I'll probably pour a set of (nearly) nuteless agar plates to minimize the tetracycline use. That stuff is hard to come by here, so I keep a tight rationing on those capsules...
I could imagine it not working perfectly, maybe the bacteria can just feed off the mycelium's reserves, and hitch along through even the tetracycline because of the intertwining. If that's the case, the hot pour would probably be my best bet, maybe even a set of subsequent hot pours.... Oh well, we'll see..
