Tahnkyou thankyou thankyou for all of your hard work. Whenever i want to learn something from books/web, i want to find trusted sources who are not afraid to question silly, convoluted, outdated practices. And collect simple, elegant, and practical solutions . Your work is just that! Like that stuff with the hygrometer! of course so obvious, and many other things. Subarashii!

Would grains that were sterilized way too long still be useful? Moisture content looks okay but they are darker than average after the cycle. PC didn’t run dry or anything- just concerned about the bioavailability of the nutrients and whether or not the oats will still be just as easily colonized.

Are you using a speed controller with your flow hood? I'm assembling mine now and have the same fan as you for a 2x2 cabinet. I bought the controller most on here use and realize its not intended for use with these motors. I'm seeing a lot of suggestions to start the power at max and then back off until I reach 100CFM. I think I'd prefer that than covering part of the intake so that I can scrub at high speed and dial it up easier as the filter gets a bit more congested. Thanks.

The dayton motor I use shouldn't be used with a speed controller. (A frequency drive would work tho $$$$) so i block the intake. The motor runs at full power but I can remove my restrictor plate for scrubbing then slap it back over the intake to throttle it to 80-120fpm

I sterilized grains overnight once. They didn't work. Ymmv

Syringes are inherently dirty by nature. Those look like they're carrying a lot more than just cube spores. The plate on the left in the right pic looks like you might be able to get a small piece of clean growth to transfer but there's a ton of other stuff hanging out. I'd personally use the syringes to inoculate some brf cakes and then clone a fruit from that rather than try to clean that up, especially if you say you have had success streaking plates with spores

Yeah, i was figuring that as well, just want to confirm (pretty obvious its not cube).
Im not going to try and extract that little guy in there, I just want to get some cubes that grow.
I inoculated some rye jars, only one has a little mycelium starting to go, the other three show nothing after a week.
Any good sources for cube prints on here that ship to Cali? I cant find anyone that ships here.

I had some MS syringes of Ecuador that looked just like your pic to the right...after about three weeks they germinated to nice plates for transfer. I've found two things that help...first the advice of BRF is spot on! It is very forgiving and the mycelium beats the contams of syringe 9/10 times, then you can clone best phenotypical specimens. The other thing that has helped my MS to agar is stop streaking the plate. We do that in the lab with isolated cell cultures to have multiple colonization points to study. If you place a drop center plate, the mycelium will generally beat out the contamination and then you can find more uniform edges to transfer the cleanest, most uniform, growth away from.

 T1 from above plate
The plates above were 2 drops a little off center because I had a pretty 'foggy' SAB that day so my aim was off, but you can at least see how without the streaks they provided better edges to choose from. Good luck Holmes! HH

That is bacteria. Don't put nearly so much spore solution on the dish. Like one drop is more than enough

Work on finding prints on the trade list. I've recently received a wicked clean print from a variety I've been wanting to track down for awhile. When you are reading through post look for signatures that have 'trade list' in em and then pm the poster. You'll find dem prints last forever and are much better than MS syringes from any vendor typically. hh

I did put a lot less (one drop) on this latest run though and the bacteria is forming again :/
It forms way before any of the mycelium take hold. Im thinking I just got burned and will look for prints here. I have pan cyan spore prints that are working out really great so far, working on getting to monoculture.

Why is leucism a desired trait? I've seen several attempts to gain leucistic fruit, but I'm unsure why. Does it affect the growth or potency of the mushroom? Novelty?

Hello, my question is simple and I'm sorry if it was answered before, but I can't find definite answer using search. Do you spray monotube after pinning started ?
