film on agar
    #2441613 - 03/17/04 01:29 AM (13 years, 5 months ago)

I have got this film on all of the agar plates. The agar is completely set, no condensation on the lid but is this just water?

When you tilt it, you can see the liquid run down. The spores have germinated so I am not too concerned, but is there any way to avoid it?

If it's not contaminated then it's caused by the water that condensates on the agar surface after the agar solidifies and IME it depends from the material used for wrapping. I tested 3 types of wrapping:
micropore medical tape - no condensation, but cultures dried quickly
polyethylene wrap - lots of condensation, probably doesn't allow much air exchange
parafilm - far the best. It seems expensive at first look, but it's very strechable, so you can use 1 roll for lots of petris. I'll never try anything else.

They aren't contaminated as far as I can tell. Nothing nasty. May have to try that parafilm and see how these current cultures turn out in the meantime.

thanks for the info. cheers!

Try this method next time to prevent condensation at pouring:

I found Anno's link very helpful. Another suggestion: Flip the petri dishes so that the side with the condensation is at the bottom (agar on top). I found that this helps with the condensation within a day or two.

Thanks. I tried doing it that way and my water in the mugs must not have been hot enough. I think I got it figured out now. Latest plates look really good.

cheers!
