Initiate fruiting
    #24409803 - 06/16/17 10:51 AM (3 years, 10 months ago)

Hey guys, Ive gotten to the point where I have a monotub full of cultured grain. Im growing on wheat and the strain is "Z" My smaller tubs have gone grey overnight. How do I initiate fruiting other than lowering temp and humidity? Also, are the smaller tubs saveable?

Trash them, trash them now.

Good god thats really bad.

Do ypu have any pics of youe jars? How did you start? Did you PC?

I started spore syringes on agar and produced a liquid culture from that. I innoculted cooked, sterile wheat (I understand sterility as I body pierce and have repurposed an autoclave to do my sterilizing) with my LC and incubated at 84° until 80% colonized. I know my agar cultures are DEFINITELY mycelium because a tiny mushroom sprouted from it. Not being cheeky, but what makes you say its trich? The pic is bad but its mostly pure white.Ive reasearched the SHIT out of this difficult to navigate website but OBVIOUSLY still dont get it. I thought a guy could fruit grain without coir casing? Isnt Vermiculite only necessary if humidity is difficult to control? PLEASE HELP! Info and links would be greatly appreciated!  Here is a better pic. Not Mycellium? Nm cant fucking load pics for some dumb fucking reason. I guess ease of use is second rate.

Ok, thats what I figured when I opened agar jars and they smelled like fresh cut store mushrooms. So how do I get trich. on the grain? Humidity too high? Temp too high?

The trich was already on the grains, it could've been your grain prep or the syringe or your inoculation.
There was a mistake made somewhere.

Describe from start to finish how u did it.

Im thinking contaminated LC? Is it possible to grow a liquid culture of Trich? I didnt add h2o2 after it cultured, maybe that was trich and not mycellium? I opened the LC and it does not smell. How could I tell if the LC is contaminated? I will describe the methods I used when I get a bit to write it up.

Did u clean up your culture with agar or put spores in to start lc?

I took a chunk of agar culture and put it into my sterilized dextrose/water medium.used sterile tools and dipped in alcohol and burned between each cutting i took. I made three. I scrapped one that looked like yellowish culture. One i added h2o2 and the last I left as is. I inoculated 1 jar with each liquid culture I had left. The one with h2o2 grew nothing so I spawned the other culture to grain jars thinking I added too much h2o2. I used 2 ml of 3volume diluted with 7ml water and added to 960ml culture.

I dont think you should be working with LC until you are more experienced.
84° F is way too hot for colonization BTW. Try closer to 70°.

If you have clean, fully colonized grain spawn yes you can fruit it by itself in trays although typically you would case it.
Its much better to spawn to a bulk su strate, such as coir, by mixing your colonized grain spawn (at ratio) into the coir evenly.

I suggest you start over by inoculating a new agar culture and make transfers until you are confident it is clean. Skip the LC. Inoculate the grain directly with agar wedge and expand via grain to grain culture (or inoculate all your grain spawn directly with agar wedge).

Also, did you test your LC before using it? *Always* have to test it on agar.

Lose the peroxide too. It has no use in mushroom cultivation.

Hydrogen peroxide? You dont use that ever.

You transfer 3 more times after u think its clean as a rule of thumb.
Your culture was either dirty or it got in when inocd your lc.
Did u use a SAB?
You've read some really bad info, research that link i gave u, use the info on this site under thats under 5 years.

Thanks guys. I understand the bulk substrate thing a little better now after reading a few of the links provided and will definitely switch to agar wedge transfers. Just so I get this straight; transfer mycellium grown from spore syringes 3× agar to agar to guarantee isolation? Then transfer agar to grain jar and fully colonize, then mix grain with coir and vermiculite (sometimes gypsum) in monotub and colonize again, THEN initiate fruiting?

Its not isolatation its just a rule of thumb for clean culture after u think you've already got clean cultures.
But for the most part yea, post your jars here so some1 can give u the thumbs up/ thumbs down on your myc.
An post again when before you spawn so the pros can tell you whatsup

Yep first thing is to get yourself a clean culture. Here are a couple examples on a PP5 plate and a pitre.
It takes however many transfers it takes until you have healthy myc and there is no bacteria or mold colonies present on the plate.
You use this to inoculate your grain.
You can either use this to spawn or expand it to more grain (G2G transfer).

Then you can either case the grain and fruit it , or (much better) spawn it to a bulk substrate and fruit it. Casing is optional for Cubensis.

I'd suggest starting small, like each grain jar you get of fully colonized grain spawned to your substrate and put into small totes. Get the hang of that first and then move up in size  :2cents:
