does agar mean higher yeilds?
    #24404327 - 06/14/17 08:40 AM (3 years, 10 months ago)

I'm trying pasty plates again and I'm trying to understand why or if agar to wbs can result in a higher yielding tub versus just innoculating with an ms syringe.

First of all, working with agar allows you to eliminate contaminations.
Then you can drop healthy myc into jars with grains, let them colonize fully and you can spawn to bulk which means higher yield in general.

Usually inoculating with syringe leads to contamination.

Yes.
Less contams, isolated genetics, clear growth, and fast, and easy to expand all give you more Fruits!

Why are syringes associated with contams? I mean, I've experienced it. I just lost two out of the three tubs I prepped for bulk grows and it fucking sucked.

All of the jars were from homemade syringes, in which I had good sterile technique setting up.

I guess I don't see how dropping spores into distilled water and sucking it into a syringe is anymore sterile than dropping some spores onto a plate and watching it colonize. I don't have a lot of experience with the plates really. I did petris and they all contamed.

You move over the clean part to another agar plate until you are contam free.
Syringes, you cannot isolate good growth.

Chances are higher for success with agar.

Agar is used customarily as a gelling for nutrients so you can quickly inoculate a bunch of petri plates and have a bunch sitting around to make transfers to.
The question of better growth and bigger flushes is a good one but I have never used anything but small white rice jars for transferring good growth and finally adding that to whole grain jars to get my main spawn for doing bulk grows.
I would say agar would be nice if I had a complete set-up for growing but ghetto for now is what I have to work with and my flushes go toe to toe with most in weight category.

Most cube varieties are pretty stable I personally thing focusing on dialling in conditions is a first for newer growers and agar to make sure what your using is clean

Okay I made 8 plates. I can't post the pics, but I'm confident in my agar mix. I put spores in each with a flame sterilized exacto and did all of it in a sab.

How long should this take to have something I can dump in a jar?

Well if you're contam free probably about a week, but idk for sure because I don't use spores on plates, I use live spawn

few days to weeks and you should see growth

i prefer to streak plates with a serial streak ,as described in my agar guide, since it makes it so fast and east to isolate or clean a culture, with hundreds of initial colonies of various genetic diversity to choose from

But in any case you won't want to inoculate with the spore plate for these exact same reasons. even if there arent visible contams on the initial plate, something might show up when it hits grain. for this reason (among others) you should do a few transfers first to ensure you have a clean, well-organized culture, then use that to inoc

agar (used appropriately) means way fewer contams (if any), and it also gives you the ability to clone and isolate, which are the key to consistency

Three of my right plates are showing healthy non contaminated growth. They're going to get at least another week chilling on the shelf time.

My question is can I dump a healthy one into a wbs jar?

The other two I was going to splice out into some other plates to see what they do. Is it strange to see plates go green like this? I was as sterile as I could be and used a sab.

Agar is the mushroom equilivent of artifical selection. U can pick and choose what cultures to grow, what fruits to clone, elminate infections.

All this leads to higher, more consistent yields.

Makes sense.

So the three cultures that look good... 25 percent of the surface area of the plate is white and fluffy and strong looking. Is it about time to dump it into a clean wbs jar??

Once you find a clean culture youll want to transfer about 3 more times to ensure its health. Just because it looks clean doesnt mean it is best make sure by cutting out tbe best bit (fastest growing cleanest) and sticking it on a new plate then repeat. Never use the very first plate thats no different than just using the spore syringe as is into the grains.
Also stick up pics and we can help you choose the sections to transfer from.

Yeah, you usually want to make at least one transfer before you inoculate grains... a little patience will go a long way.
