First time with agar
    #24342170 - 05/23/17 05:31 AM (6 years, 10 months ago)

MS of ATL7

I'm thinking of transferring the section of growth in the middle of the first picture. First picture things look mostly good to my newb/untrained eye. Not sure what's going on in second picture. I would love to hear what those who know more than me think of

I've only done PF and MS to grain before. So I'm thinking once the clean culture grows out part of it will go into a G2G master jar and another part in a LC jar and see which method I like the best.

Looks good to me! Second pic looks like some bacteria has hitched a ride with the mycelium.
+5 for starting agar

first pic looks better, transfer from the fastest clean growth, which seems so be the largest unless it was there long before the rest. Transfer from the leading edge, and it doesn't need to be a hunk of a chunk. Nice looking growth btw!

On the center area of that 1st pic, I'd take transfers from about 4 o'clock to about 9 o'clock.

2nd pic - center area also from the top edge.

If you are using 2" parafilm tape, cut that in half length ways to make 1" strips. 3 squares long.

Yup I like the 1st plate better. Transfer piece should be about the size of a grain of rice, from the leading edge.

Why would you take the transfers from 4 and 9 o'clock?

Both plates were setup at the same time, so the large piece is definitely the fastest. Was very happy it grew out like that!
End goal for this is a good sclerotia producing strain.

Thanks for the info guys

it all looks good to transfer from, both plates. good job!

you will find that spores germinate at different times and match up with other compatible germinated spores at different rates (and in different quantities) and get different looking mycelium from multispore. its no problem at all. nothing there looks like definitive contams.

one suggestion tho, if that is parafilm wrapping those plates stretch it out as you wrap it to make a nice tight seal. you will snap it a few times to figure out how much to stretch it but thats ok. even if its not parafilm you can get a much better seal stretching it as you wrap those dishes.

good luck and keep up the good work

First two transfers done, took from the fastest growth in the first pic. I am going to let the second grow out some and see if any stones form.

Thanks for the advice on cutting the parafilm in half and stretching it, it definitely made a difference!

Argh, I got some mold :frown:

Here is the other one:

The mycelium (or maybe mold) on the agar is growing upwards, is this normal or is that mold?

Should I trash the first one, or let it grow out a bit and transfer?
