my cultures have been shit lately.
 
i confess its really bugging me. i can deal with satellite contams from poor technique or just dirty as spores but this weak ass bacterial shit is tripping yo.
multiple varieties, multiple syringes.. prints.. something im doing is fucked.

This has been my struggle for a while now. I hate that shit. I recently made a couple transfers from a plate like that useing an inoculation loop to snag a bit of mycelium. Will see if that helps.

i think i really fucked up the agar..but all my growth is shit. ill pour new plates today and transfer some things just to double check but im tossing like eeeeeverything from this batch. luckily is not too many dishes. maybe 30.
i also broke down and ordered 100g of dehydrated pda and mea.. the wife wants to make potato salad so and im soaking some oats right now so ill save that water for later uses also.

i mean i messed up th batch of agar.. when i made it. i poured great!
but i fuckd up the batch of agar. like i cut into a plate and the the media had a skin and underthat was agar goop.
no bueno.
my scalpel flames like a homo. i use gloves.. sab(been using for while now) mist towel rack

That's strange haven't had that happen yet.
I flame my entire xacto knife handle. Like the entire thing. I'll take a picture in a sec if I remember. Once I started doing it my bacteria problems disappeared on agar.

i opened a brand new sterile disposable scalpel for that session that gave me the shit show.
it was almost the 'cleanest' attempt ive done. new blade, fresh plates, i even did a weekly wipe down of the sab with iso and lysol wipes just to clean up slopped grains and shit.
i feel like i isolated mold/bacteria with the transfers. oh well.
all you can do is make more shit and try again

i have! i got the tip of a atl7 swab cut off and buried in a plate thats about to grow on the surface. should be cool. and ive got a sandwich going on with some pans if been struggling with.

i dont feel like going through all that for a random cube though.

I do just the opposite, everything sketchy gets tossed, provided I have a clean(er) culture of the same "strain" going alongside it. That's why 90% of the time, I make 2-5 plates every time I actually only need 1. Contams on agar tend to scare me. I'd rather go around :p

Grrrrr.. I'm really starting to hate my PC right about now...

I angle-grinded my trivet legs off so I could fit more PP's in my PC, after that I filled it with more PP's than it actually had room for, and then I ran it dry in just over an hour at pressure, leading to warped upper PP's, a plastic covered trivet, some rims of what used to be the lower PP's, and a PC bottom with a plastigrain mix.
FUCK. THIS. SHIT. I need me a new, decent PC, that won't run dry like an Ethiopian.
